Potenzial adipogener Stammzellen bei radiogenen Wundheilungsstörungen

Abstract: The increased expression of cytokines and adhesion molecules by NHF and HDMEC after external radiation is mitigated in the co-culture setting with ASC. These in vitro changes seem to support the clinical observation that ASC may have a stabilizing effect when injected into irradiated wounds.

“…In addition, we observed increased amounts of VEGF after external radiation of NHF and ASC mono-cultures in previous studies [ 28 ]. Irradiation and surgery are well known to induce hypoxia [ 32 ] which is a relevant factor in inducing VEGF transcription [ 33 ].…”

“…Comparable to the effect of external radiation on HDMECs, we found modulatory effects of ASCs concerning the IL-6 expression in NHFs [ 28 ]. IL-6 is a potent inducer of MMP1 and TIMP1 in dermal fibroblasts [ 29 ] and has also been reported to increase MMP13 expression in human chondrosarcoma cells by Tang et al [ 30 ].…”

“…Alone, or combined with scaffold, ADSCs have proven efficiency in restoring maxillary defects, accompanied with apparent bone regeneration [164]. Post-traumatic calvarial defects were also treated with autologous ADSCs and calvarial continuity with bone regeneration were obtained [165]. Their ability to secrete anti-inflammatory factors and decrease pro-inflammatory responses also makes them very attractive and efficient in the treatment of inter-vertebral disc damages, rheumatoid arthritis, osteoarthritis, and tendon damages [19,22,150,151,152,153,154].…”

“…Mesenchymal stem cells are suggested to support wound healing due to their production of multiple growth factors and cytokines which are of major interest in wound healing processes [ 4 ]. The discovery of adipose-derived stem cells (ASC) also as multipotent progenitors of various cell types was an important step forward in the field of stem cell research [ 5 , 6 ]. Furthermore, ASC can be obtained easily by rather low invasive procedures such as liposuction.…”
