− The effects of Picrorrhiza Rhizoma (PR) aqueous extracts were observed on xylene-induced acute inflammation. The xylene was topically applied 60 min after administration of 500, 250 and 125 mg/kg of PR extracts, and all animals were sacrificed 2 hrs after xylene application. The changes on ear weights, histolopathological analyses of ear were evaluated as compared with indomethacin and dexamethasone 15 mg/kg treated groups -well known anti-inflammatory agents. Xylene application resulted in marked increases in induced ear weights as compared with intact control ear. Severe vasodilation, edematous changes of ear skin and increase in the thickness of the ear tissues as acute inflammation were detected in xylene-treated control ears at histopathological observation. However, these xylene-induced acute inflammatory changes were dosedependently decreased by oral treatment of PR extracts. Therefore, it is concluded that PR extracts has favorable anti-inflammatory effects on xylene-applicated acute ear inflamed mice.

Abstract.[Purpose] The purpose of the present study was to investigate the effect of magnetic infrared laser (MIL) radiation on the xylene-induced acute inflammation.[Subjects] The subject animals were male ICR mice.[Methods] Mice were once irradiated (60s) by MIL at 1.33, 2.66 or 6.65 J/cm 2 of MIL, or treated with indomethacin or dexamethasone. Then xylene (0.03 ml) was topically applied to the anterior surface of the right ear to induced inflammation 1 h after irradiation with MIL. The changes in ear weights, histological profiles and histomorphometrical measurements of the ear were conducted upon sacrifice. All animals were sacrificed 2 h after xylene application. Results of MIL irradiation were compared to those of indomethacin and dexamethasone (15 mg/kg injected once intraperitoneally).[Results] Xylene application resulted in marked increases in xylene-insulted ear weights compared to that of the intact control. Thus, the differences between intact and insulted ears were also significantly increased. The histological characteristics of acute inflammation, such as severe vasodilation and edematous changes of skin, were detected in xylene-treated control ears with marked increase in the thickness of the ear tissues. However, these xylene-induced acute inflammatory changes were significantly and dose-dependently decreased by MIL irradiation. [Conclusion] We conclude that MIL therapy has a favorable effect in the reduction of the acute inflammatory responses induced by xylene application in mice.

(2005 The emphasis of the present study was to investigate the relationship between inducible nitric oxide synthase (iNOS), RNS, P-selectin and platelet-arteriolar wall rolling/adhesion and microvascular constriction in the intact lung during the time course of pulmonary ischemia-reperfusion injury.We examined the hypothesis that pulmonary IR induces platelet-arteriolar wall rolling and adhesion (interactions) via a P-selectin dependent mechanism that contributes to pulmonary microvascular constriction during reperfusion. Increased P-selectin expression results from elevated iNOS activity and subsequent RNS generation. Subpleural arterioles, labeled platelets, and leukocytes were examinedin anesthetized, open-chest rabbits by intravital fluorescence microscopy. Ischemia was caused by reversibly occluding the right pulmonary artery for 5 min, 1 h or 2 h (5minIR group, lIR group and 2IR group, respectively).During reperfusion, postischemic platelet rolling and adhesion were independent from leukocyte-arteriolar wall interactions and correlated with pulmonary arteriolar constriction in proportion to the length of ischemia. After l-h occlusion during reperfusion, platelet rolling was significantly greater than in the 5minIR or control group. Although arteriolar diameters decreased during the first 0.5 h of VI reperfusion, platelet adhesion was not increased and arteriolar diameters returned to the baseline by 1 h of reperfusion. However, after two hours of ischemia, platelet rolling increased and platelet adhesion was accompanied by arteriolar constriction that was correlated with the level of platelet adhesion after both I-h and 2-h ischemia. Blockade of systemic P-selectin by Fucoidan (the selectin ligand) inhibited platelet rolling, adherence, and vasoconstriction. Pretreatment of only exogenously labeled platelets with monoclonal antibody (MoAb) to P-selectin prevented platelet rolling and adherence, but not vasoconstriction. In rabbits that were treated with an iNOS inhibitor (1400W) before occlusion (2IR+ 1400W group), platelet-arteriolar wall interactions and vasoconstriction were prevented. In lung tissue subjected to IR, iNOS activity and expression were markedly greater than control and were also dependent on ischemia duration. Immunochemically detected P-selectin and nitrotyrosine expression in ischemic lung tissue, and the plasma level of soluble P-selectin in lIR and 2IR groups, were significantly higher than in non-ischemic lungs and were inhibited by pretreatment with 1400W.These results indicate that during reperfusion of the intact lung, pulmonary IR causes platelet rolling and adhesion along arteriolar walls and suggest that this process is mediated by platelet and endothelial P-selectin.
