Potential Role of Aromatase over Estrogen Receptor Gene Polymorphisms in Migraine Susceptibility: A Case Control Study from North India

Abstract: BackgroundThe present study was undertaken to find out the role of estrogen pathway related gene polymorphisms in susceptibility to migraine in Northern Indian population. Aromatase, CYP19A1 (rs10046 and rs4646); estrogen receptors, ESR1 (rs2234693, rs1801132, rs2228480 and rs9340799) and ESR2 (rs1271572 and rs1256049) polymorphisms were selected for the present study.Methodology/Principal FindingsThe patients were recruited in two cohorts – primary (207) and replicative (127) along with 200 healthy controls a… Show more

“…A meta-analysis of estrogen polymorphism studies indicates that two variants, ESR-1 594G> A and 325C> G, increase the risk for migraine 40-60% (99). Challenging that study, however, recent evidence indicates that aromatase polymorphisms (CYP19A1 rs10046 and CYP19A1 rs4646) also confer a risk for migraine and its protective effect, respectively, and may be more significant than estrogen polymorphisms (100). The latter hypothesis is supported by a twin study indicating that environmental and genetic factors have comparable contributions to migraine (101).…”

“…The ESR1 rs2228480 (A allele) may be associated with an increase in migraine susceptibility in Caucasian populations, and it was also associated with increased coronary intima media thickness in women, suggesting an increased susceptibility to cardiovascular disease . A link between migraine and the rs10046 and rs4646 polymorphisms in the estrogen producing enzyme CYP19A1 has been demonstrated …”

“…However, the extensive research fail to reach a consensus with respect to the inherent susceptibility to migraine associated with ESR1 gene polymorphisms. 16 21 The small numbers and varying populations of the published studies may partially account for the controversial results. The most important reason that promotes us to perform the present meta-analysis is the less reliable discoveries of an earlier analysis as a result of overlapped data 22 and the new information from recent publications.…”
