Potential Preventive Strategies for Amyotrophic Lateral Sclerosis

Abstract: It may seem useless to propose preventive measures for a disease without established pathogenesis and successful therapy, such as amyotrophic lateral sclerosis (ALS). However, we will show that ALS shares essential molecular mechanisms with aging and that established anti-aging strategies, such as healthy diet or individually adjusted exercise, may be successfully applied to ameliorate the condition of ALS patients. These strategies might be applied for prevention if persons at ALS risk could be identified ear… Show more

“…A positive correlation of plasma leptin and body mass index (BMI) was observed in ALS patients (Ngo et al., 2015). This is of interest because patients with ALS are unable to maintain their body weight (Bouteloup et al., 2009; Desport et al., 2001), and rapid weight loss is clinically associated with worse disease outcomes (Ahmed et al., 2019; Kuraszkiewicz et al., 2020). Indeed, weight loss is a predictor of shorter survival in ALS (Lee et al., 2021), raising the possibility that leptin could be exploited to derive therapeutic effects.…”

“…Various treatments are used to alleviate symptoms but there are only two pharmacologic agents indicated for the supervision of ALS. Riluzole in the year 1996 had Copyright © 2021 IR Research Publication -All Rights Reserved been the anchor for disease-modifying therapy, but in May 2017, a new agent Edaravone is approved by the FDA [11,14,16]. Various organizations are dedicated to work for multidisciplinary care by imparting aid for people with ALS-support, ALS Research and clinical trial, therapeutic progression for irregular and congenital forms of ALS.…”

“…Furthermore, other factors including oxidative stress, mitochondrial dysfunction, and neuroinflammation are implicated in ALS pathologically. Neuroinflammation, via NF-κβ proinflammatory cellular pathway activation, plays a central role in ALS pathogenesis [13,59,60]. Several ALS patients had been shown an increased NF-κβ signaling expression in microglia.…”

“…Several ALS patients had been shown an increased NF-κβ signaling expression in microglia. Thus, through NF-κβ expression reducing in microglia can slow the progress of disease and lengthen the survival in ALS mouse models [60,61].…”
