Potential Mechanisms Connecting Purine Metabolism and Cancer Therapy

Abstract: Unrestricted cell proliferation is a hallmark of cancer. Purines are basic components of nucleotides in cell proliferation, thus impaired purine metabolism is associated with the progression of cancer. The de novo biosynthesis of purine depends on six enzymes to catalyze the conversion of phosphoribosylpyrophosphate to inosine 5′-monophosphate. These enzymes cluster around mitochondria and microtubules to form purinosome, which is a multi-enzyme complex involved in de novo purine biosynthesis and purine nucleo… Show more

“…Hence, we have titrated only the amino-group, not the phosphate group. To titrate the GAR ligand, we have computed the restrained electrostatic potential (RESP) charges 53,54 in both protonated and deprotonated states (Table S1) and the reference energy ( 0809,/01 ) as defined in equation- (1).…”

“…Gene function (GO biological process and KEGG pathways) annotation revealed that the two bad survival lncRNAs (FIRRE and LINC00839) of the four‐lncRNA signature are involved in the processes, which promote cancer progression such as telomere maintenance, DNA replication, and DNA repair. It seems that metabolism processes also play an important role in NB since the function prediction identified many metabolism processes such as purine metabolism, tRNA metabolic process, nucleotide metabolic process, cysteine, methionine metabolism, etc In fact, purines are basic components of nucleotides in cell proliferation, thus, impaired purine metabolism is associated with the progression of cancer 44 . However, how does purine metabolism affect NB progression or regression is unknown.…”
