Potential lipolytic regulators derived from natural products as effective approaches to treat obesity

Abstract: Epidemic obesity is contributing to increases in the prevalence of obesity-related metabolic diseases and has, therefore, become an important public health problem. Adipose tissue is a vital energy storage organ that regulates whole-body energy metabolism. Triglyceride degradation in adipocytes is called lipolysis. It is closely tied to obesity and the metabolic disorders associated with it. Various natural products such as flavonoids, alkaloids, and terpenoids regulate lipolysis and can promote weight loss or… Show more

“…Existing studies have also demonstrated that flavonoids can regulate lipolysis, promoting weight loss and improving metabolic conditions associated with obesity (X. Yang et al, 2022). Flavonoids have been linked to preventing weight regain after successful weight loss (Lundgren & Thaiss, 2020).…”

“…In addition, capsaicin chronically activates TRPV1‐induced Ca 2+ influx and induces the “browning” phenotype in 3T3‐L1‐adipocytes 82,107 . The activation of TRPV1 by dietary capsaicin increases CX43‐mediated Ca 2+ influx in adipocytes, thus promoting adipocyte differentiation 68,108 . It has been shown that TRPV2 knockout reduces Ca 2+ influx in BAT and weakens β‐adrenergic receptor‐stimulated thermogenesis 70,109 .…”

“…In an oversimplified scenario, the activation of lipolysis would represent a suitable treatment for overweight/obesity. However, experimental evidence shows the lack of lipolysis enzymes (such as adipose triglyceride lipase, hormone sensitive lipase, or lipase monoglyceride) in a nonobese or only moderately obese phenotype in humans and mice, while highlighting an increase in basal lipolysis in obesity (Yang, Ge, et al, 2022). The excessive release of free fatty acids may damage tissues, contribute to the progression of obesity, and promote obesity-associated metabolic complications, such as insulin resistance.…”
