Potential application of urea-derived herbicides as cytokinins in plant tissue culture

Abstract: Various urea-derived herbicides and different cytokinin analogues were used to determine their effects on callusing response and shoot regenerating capacity of alfalfa (Medicago sativa L.) and Coleus (Coleus forskohlii Briq.). The herbicides monuron and diuron evoked profuse callusing response from Coleus leaf segments and alfalfa petiole explants on Murashige and Skoog medium. Shoot regeneration by monuron (2.0 mg/l) showed a maximum of 3 multiple shoots both in alfalfa and Coleus with a frequency of 92% and … Show more

“…Some PGRs are herbicides, including 2,4-D, TDZ, picloram and dicamba, are popular in plant tissue culture studies, and when added to medium at high concentrations, may kill plant tissues and cause developmental aberrations. TDZ is traditionally used as a herbicide in agriculture but became increasingly employed in plant tissue culture 17 . TDZ affects plant physiology, such as cellular, energy, nutrient, transport and metabolic alterations to the membrane, and in flowering Dendrobium , alters the endogenous levels of PGRs 18 .…”

“…Cytokinins play a central role in the regeneration of multiple shoots in many medicinal plant species [32,33] and sometimes in combination with auxins. Cytokinins trigger cell division and influence differentiation [34], while auxins are a major signal for apical dominance [35] and are responsible for elongation in phototropism and gravitropism [36]. In vitro plantlets will then be transferred to potting media during acclimatization, the final stage.…”
