Potential anticancer effects of cell wall protein fractions from Lactobacillus paracasei on human intestinal Caco‐2 cell line

Abstract: Consumption of probiotics has an important role in colorectal cancer prevention. In this study, we aimed to explore that the cell wall protein fractions from Lactobacillus paracasei could induce apoptosis on Caco‐2 cell line. The cell wall proteins from L. paracasei were fractionated by gel filtration chromatography (F1, F2 and F3) and characterized by polyacrylamide gel electrophoresis (SDS‐PAGE). The anticancer properties were evaluated using MTT assay and Annexin V‐FITC/PI staining. Administration of L. par… Show more

“…Due to the different mechanism of action, several publications applied both methods to investigate cytotoxicity of different chemicals [28][29][30]. In order to further study cytotoxic properties of PEGs, we used labelled annexin V and propidium iodide to discriminate between necrotic and early apoptotic cells with flow cytometry [29,31,32]. Autophagosome formation was measured by cellular organelle staining as a marker of autophagy, an inducible mechanism of cytotoxicity [33][34][35].…”

“…Heat-treated cells, crude cell walls and intracellular extracts from Lactobacillus casei 01 can significantly reduce the activity of HT-29 ( p < 0.05) and play an anti-proliferation role [ 40 ]. A study showed that cell wall protein fractions from Lactobacillus paracasei can inhibit the proliferation of the human colon cancer cell line Caco-2 and promote apoptosis [ 41 ]. Heat-inactivated probiotics can promote the production of Th1-related cytokines and reduce Th2-related cytokines to participate in regulating the immune system [ 42 ].…”
