Posttranslational Protein Modification in the Salivary Glands of Sjögren’s Syndrome Patients

Abstract: The present study investigated posttranslational reactions in the salivary glands of patients with Sjögren's syndrome. We analysed the biopsies of primary Sjögren's patients using immunohistochemistry and a tag-purified anticyclic citrullinated protein (CCP) antibody to detect citrullinated peptides, and the presence of peptidylarginine deiminase 2 (PAD2) was assessed simultaneously. The present work demonstrated the weak presence of the PAD2 enzyme in some normal salivary glands, although PAD2 expression was … Show more

“…Citrullination refers to the post-translational conversion of protein arginine residues into citrulline residues, a process catalysed by peptidylarginine deiminases (PADs) 1–4 and PAD6 [ 1 ]. Citrullination plays an important pathogenic role in anti-citrullinated protein antibody (ACPA)-positive rheumatoid arthritis (RA) [ 2 , 3 ] and, possibly, in a number of other autoimmune diseases, inflammatory diseases or neurodegenerative conditions, including multiple sclerosis [ 4 ], Alzheimer’s disease [ 5 ], psoriasis [ 6 ], Sjögren’s syndrome [ 7 ], type 1 diabetes [ 8 ] and chronic obstructive pulmonary disease [ 9 ].…”

“…Previous studies have reported PTM (post-translational modification)-modified proteins in patients with pSS such as citrullinated Ro60 and histone 1 ( 89 ). Furthermore, salivary antibodies against cyclic citrullinated peptide (anti-CCP), a proxy for anti-citrullinated protein antibodies (ACPA), have been identified in patients with pSS ( 90 ), suggesting that citrullinated proteins can be a utilized as diagnostic biomarkers. “…One study investigated the influence of inflammation from periodontal disease using RA-model mice analysis in serum and saliva samples of both anti-citrullinated proteins (using antibody analysis) and citrullinated proteins (using gel electrophoresis) [ 180 ]. Saliva and serum showed similar results, evidencing protein citrullination around the 55 kDa molecular-weight electrophoretic band.…”

“…Patients affected by Sjögren’s syndrome have been found to be positive for ACPAs and showed increased levels of the PAD 2 enzyme, suggesting a possible role of citrullination in the disease that could be responsive to autoantigens triggering [ 180 ]. However, in the same study, it was outlined that only approximately 7.5% of Sjögren’s syndrome patients are positive for ACPAs.…”
