Postendocytotic sorting of the ligand for the polymeric immunoglobulin receptor in Madin-Darby canine kidney cells.

Abstract: Abstract. The polymeric immunoglobulin receptor (pig-R) is responsible for the receptor-mediated transcytosis of polymeric immunoglobulins (IgA and IgM) across various epithelia. We have expressed the cDNA for the plg-R in Madin-Darby canine kidney (MDCK) cells and found that this system mimics that found in vivo (Mostov, K. E., and D. L. Deitcher. 1986. Cell. 46:613-621). We have now investigated the postendocytotic pathway of the ligand for the plg-R. After a 5-min internalization at the basolateral surface… Show more

“…The postendocytotic fate of a preinternalized cohort of 125 I-IgA (at 5-10 g/ml) was analyzed as described (40). In brief, filtergrown MDCK cells expressing the wild-type pIgR were treated with or without drug, and 125 I-IgA internalized from the basolateral cell surface of the cells for 10 min at 37°C.…”

“…Endocytosis of 125 I-IgA was measured as described (40). At the conclusion of the experiment, 125 I-IgA was stripped from the cell surface by incubating the cells for 60 min at 4°C with 750 mM glycine, pH 2.5, diluted 1:5 with PBS ϩ (Dulbecco's phosphate-buffered saline with 0.5 mM MgCl 2 and 0.9 mM CaCl 2 ).…”

“…With the notable exceptions of NgCAM, and polyimmunoglobulin receptor, apical proteins are likewise delivered to the apical plasma membrane without first being delivered to the basolateral surface (Breitfeld et al, 1989, Anderson et al, 2005Hua et al, 2006;Paladino et al, 2006). Delivery to apical and basolateral domains is via separate transport vesicles Sztul et al, 1991).…”
