Possible Influence of Glutathione S -Transferase GSTT1 Null Genotype on Age of Onset of Sporadic Colorectal Adenocarcinoma

Abstract: The results suggest that inherited absence of this carcinogen detoxification pathway may not be associated with sporadic colorectal adenocarcinoma in the present cases. However, a higher frequency of GSTT1 null genotype in patients diagnosed before the age of 60 years suggests that this genotype could influence the age of disease onset in Brazil.

“…We found no association between GSTT1 status and CRC. A number of studies have shown the GSTT1 null genotype to be associated with increased CRC [4,20,21,22,23]. However, other studies have failed to replicate this association [24,25,26,27,28].…”

“…Theoretically early onset of lung cancer may have well built genetic component in its pathology than is the case for lung cancer at older ages, and many studies have reported GST gene polymorphism have strongly associated with cancer risk at younger ages (16)(17)(18).…”

“…Differential expression from the drug metabolism gene array includes functional gene groups related to Drug transporter such as metallothioneins and P-glycoprotein family; Phase I metabolizing enzymes such as cytochrome P450 (CYPs) and Phase II metabolizing enzymes such as acetyltransferases, epoxide hydrolases, glutathione Stransferases (GSTs), methyltransferases, sulfotransferases and UDP glycosyltransferases (14)(15)(16)(17)(18). Metallothionein (MT) is a sulfhydryl-and cysteine-rich protein found in microorganisms, plants and all invertebrate and vertebrate animals (14).…”

“…Mammalian cytosolic GSTs form a super family consisting of four distinct families, named α, μ, π and θ. The μ (GSTM1) and θ (GSTT1) members of the GST multigene family are candidate cancer susceptibility genes because of their ability to regulate the conjugation of carcinogenic compounds to excretable hydrophilic metabolites (16)(17)(18). Worldwide, organophosphate insecticides result in numerous poisonings each year.…”
