Positron emission tomography-computed tomography and 4-hydroxynonenal-histidine immunohistochemistry reveal differential onset of lipid peroxidation in primary lung cancer and in pulmonary metastasis of remote malignancies

Abstract: The Aim of the study was to reveal if PET-CT analysis of primary and of secondary lung cancer could be related to the onset of lipid peroxidation in cancer and in surrounding non-malignant lung tissue.MethodsNineteen patients with primary lung cancer and seventeen patients with pulmonary metastasis were involved in the study. Their lungs were analyzed by PET-CT scanning before radical surgical removal of the cancer. Specific immunohistochemistry for the major bioactive marker of lipid peroxidation, 4-hydroxyno… Show more

“…In support of this is the recent finding that 4-HNE production is increased in non-malignant cells in the vicinity of human hepatocellular carcinoma or lung metastases of remote cancer. This highlights the possible role of 4-HNE as a natural anti-cancer substance as well as a detrimental factor in (neuro) degenerative and inflammatory processes [64][65][66][67][68]. Finally, the quantities of 4-HNE protein adducts are of high importance for the cytotoxic/apoptotic effects of 4-HNE since they represent a reservoir for the persistent presence of 4-HNE, even in the absence of oxidative stress [65,69].…”

“…Such an increased resistance to oxidative stress could be one of the major advantages acquired by the transforming cells allowing them more efficient bypass of ROS damage while also giving them an important leverage compared to normal cells in the fighting chances for survival [45] . On the other hand, cancer invasion as well as inflammatory response to cancer can also induce lipid peroxidation in adjacent non-malignant tissue that can response by increased lipid peroxidation generating cytotoxic lipid peroxidation product HNE to defend themselves and the organism against invading cancer even by formation of the protein adducts of HNE [61] , [62] . Hence, it is not surprising that among common metabolic features of cancer cells is alteration of the biomembrane lipid profile by reducing PUFAs to prevent lipid peroxidation, since thus generated HNE can exert more potent cytotoxic, pro-necrotic and pro-apoptotic effects for cancer than for nonmalignant cells, also regulating activities of numerous genes, enzymes and cytokines involved in the growth regulation and oxidative homeostasis [63] , [64] .…”

“…Such a confusing findings indicating possible negative association of oxidative protein modifications and no MDA and 8-OHdG changes in PTSD indicate the need to study in particular the metabolism of HNE and the levels of HNE-His adducts in PTSD patients. Namely, the association of HNE and in particular its adducts to proteins, such as albumin, were found to be potentially very reliable biomarker of tissue specific and systemic lipid peroxidation associated with metabolic disorders, cancer and inflammatory processes including neuroborreliosis and encephalitis, which were found to be associated with systemic metabolic alterations, both reflecting like HNE-His disease progression and the efficacy of therapies used [19] , [25] , [36] , [50] , [51] , [58] , [77] , [97] .…”
