Positive Inotropic Responses of the Sodium Channel Modulator BDF 9148 in Diseased Rat Myocardium

Abstract: 1. This study has defined the positive inotropic responses to the sodium channel modulator BDF 9148 in rats with hypertension, thyroid dysfunction, diabetes or dwarfism. Concentration-response curves to BDF 9148 and calcium chloride were determined in isolated left atria and left ventricular papillary muscles. 2. BDF 9148 increased force of contraction in left ventricular papillary muscles in all disease states with maximal responses comparable to calcium chloride. BDF 9148 potency was significantly decreased … Show more

“…We have shown that there is no change in the BDF 9148 response in the early stages of hypertension‐associated hypertrophy (14‐week‐old SHR). There is also no evidence of a reduced response to BDF 9148 in left atria or left ventricular papillary muscles of 12‐week‐old SHR compared with WKY rats 8 …”

“…There is also no evidence of a reduced response to BDF 9148 in left atria or left ventricular papillary muscles of 12-week-old SHR compared with WKY rats. 8 However, the augmented responses to BDF 9148 were impaired in the later stages of hypertrophy/failure (6-18-month-old SHR). Thus, 10 ؊6 mol/L BDF 9148 prolonged the APD50 by 22 ms and augmented the contractile force by 125% in WKY rats, but only prolonged the APD50 by 12 ms and augmented the force by 87% in 6-month-old SHR left ventricle.…”

“…Hypertension causes hypertrophy of the heart and blood vessels (Diez 1992), and this hypertrophy may change the effects of drugs on these tissues (cardiac; Boyden and Jeck 1995: vascular;Kuriyama et al 1995). The positive inotropic effects of BDF 9148 are maintained in the presence of cardiac hypertrophy (Hoey, Nankervis and Brown 1995;Doggrell andNand 1995: Schwinger et al 1996). The effects of drugs on the cardiovascular system can also be altered by age.…”
