Positive feedback regulation of type I IFN genes by the IFN‐inducible transcription factor IRF‐7

Abstract: The interferon regulatory factor (IRF) family of transcription factors regulate the interferon (IFN) system, among which IRF-3 is involved in the virus-induced IFN-L L gene expression. Here we show that another member IRF-7 is critical for the IFN-K K gene induction. Unlike the IRF-3 gene, the IRF-7 gene is induced by IFNs through activation of the ISGF3 transcription factor, and IRF-7 undergoes virus-induced nuclear translocation. In cells lacking p48, an essential component of IFN stimulated gene factor 3 (I… Show more

“…4), while no such response is seen in IFNAR -/-cells. In fact, IRF7 expression is virtually extinguished in IFNAR -/-cells, consistent with known effects of constitutive type I IFN expression [35]. These data indicate that the primary mechanism of synergy does not require autocrine type I IFN signaling and up-regulation of IRF7.…”

“…IRF7 mRNA can be induced by type I IFN. The induction was suggested to be critical for IFN-␣ production through the analysis of IRF-9-deficient mice that lack IRF7 mRNA induction (27). However, wild-type DCs could produce similarly increased levels of IRF7 mRNA expression in response to 3 M ODN1668, although IFN-␣ production was profoundly decreased (Fig.…”

“…IRF7 expression is induced by type I IFNs or viral infection and involved in type I IFN production (16,26,27). Therefore, we examined whether the IRF7 mRNA is differentially induced by the two types of CpG DNAs.…”

“…4A, lane 8), implying that a STAT1-containing complex is involved in the induction of IRF-7. Because the IRF-7 promoter contains an ISRE element, it seemed to be more likely that its transcription could be induced by the type I IFNs, IFN-a and IFN-h (30). We therefore wanted to investigate the possibility that BRCA1 and IFN-g may cooperate to induce this type I signaling pathway.…”
