Positional cloning of the mouse retrovirus restriction gene Fvl

Abstract: Vertebrate evolution has taken place against a background of constant retrovirus infection, and much of the mammalian genome consists of endogenous retrovirus-like elements. Several host genes have evolved to control retrovirus replication, including Friend-virus-susceptibility-1, Fv1, on mouse chromosome 4 (refs 3, 4). The Fv1 gene acts on murine leukaemia virus at a stage after entry into the target cell but before integration and formation of the provirus. Although restriction is not absolute, Fv1 prevents … Show more

“…Fv1 is a gene on distal chromosome 4, within the linkage region mapped in this study, that has been shown to influence retroviral replication (19). Thus, Fv1 is a good candidate gene to explain the linkage data in NZ mice.…”

“…4 reverse). Additionally, the previously published polymorphic region (19) was sequenced in C57BL/10 and BALB/c mice. Briefly, PCR product was amplified with the forward primer in the presence of fluorescently labeled di-deoxynucleotides (Big-Dye terminators; Applied Biosystems, Warrington, U.K.) and purified, and the sequence was determined by capillary electrophoresis (3700 DNA Analyser; Applied Biosystems).…”

“…One of the most well characterized restriction factors is the murine Fv1 gene, which prevents infection by particular strains of murine leukemia virus (MLV) (Best et al, 1996; for review, see Bieniasz, 2004;Goff, 2004). It is known that restriction by Fv1 involves the recognition of a capsid determinant in MLV (DesGroseillers et al, 1983;Kozak and Chakraborti, 1996).…”

“…These cellular factors were first identified by Friend virus susceptibility gene 1 (Fv1), 85 a restriction factor in mouse cells that blocks specific strains of MLV. 86 Fv1 encodes a protein similar to mouse and human ERV-L endogenous retrovirus Gag 87,88 but the mechanism of Fv1 restriction remains unclear. MLV sensitivity to Fv1 restriction is determined by amino acid 110 in the viral CA protein.…”
