Portable microscopic cell counter for the determination of residual leucocytes in blood components

Abstract: The microscopic cell counter for residual WBC counting was determined to be efficient at the level of currently defined standards, with acceptable precision and accuracy. This method may prove useful for the quality assurance and control of WBC-depleted blood products.

“…The physical dimensions of the machines are (W 9 L 9 H): 220 9 375 9 250 mm. Some minor changes have been implemented in the machine compared to the one used by Bae and colleagues in their first description of the method [7]. First, the ADAM-rWBC no longer needs a laser as light source but instead uses a green LED.…”

“…Recently, a new device for the analysis of residual leucocytes in red cell concentrates was introduced [7]. The ADAM-rWBC is a microscopic cell counter measuring leucocytes on a plastic slide after fluorescent staining a blood sample with propidium iodide (PI).…”

“…In a previous report (15), this plastic microchip device was able to measure a minimum concentration of approximately 0.24 WBC/lL. The use of this platform provides an advantage over current conventional devices, as it requires very small volumes (50 lL), and the counting speed is very rapid (5 minutes/ The accuracy and precision of the new CD4 1 T cell counting system are essentially equivalent to the accuracy and precision of the dual platform flow cytometry method.…”

“…We have developed a CD4 1 T lymphocyte-counting device using a microchip and a previously described microscopic cell counter (15). This system adopts a ''nolyse, no-wash'' procedure and counts CD4…”

“…One method for the enumeration of low cell counts is microscopic counting in the Nageotte hemocytometer. This method does not require expensive equipment, but has the limitations of (i) the need of an experienced technician, (ii) a high variability and (iii) to be a time-consuming technique [1][2][3][4][5]. State of the art for enumeration of rWBCs is flow-cytometric methods with published sensitivities of approximately 0Á1 WBC/ll [3,4,[6][7][8].…”
