Porphyromonas gingivalis Gingipain-Dependently Enhances IL-33 Production in Human Gingival Epithelial Cells

Abstract: The cytokine IL-33 is constitutively expressed in epithelial cells and it augments Th2 cytokine-mediated inflammatory responses by regulating innate immune cells. We aimed to determine the role of the periodontal pathogen, Porphyromonas gingivalis, in the enhanced expression of IL-33 in human gingival epithelial cells. We detected IL-33 in inflamed gingival epithelium from patients with chronic periodontitis, and found that P. gingivalis increased IL-33 expression in the cytoplasm of human gingival epithelial … Show more

“…4A). Furthermore, IL-33 production in the culture supernatant from the cultures of the different stimulation times (12,24,72, and 96 h), which were stimulated with P. gingivalis whole cells, fimbriae, or PGTP2, also were quite low (2-5 pg/ mL), according to the ELISA findings (data not shown). Therefore, we examined intracellular IL-33 localization in BMDCs after stimulation with fimbriae or PGTP2 using immunocytochemical analysis.…”

“…Accordthelium from patients with chronic periodontitis expresses IL-33, but scarcely in that from healthy individuals (11,24). We reported previously that the contaminant related to TLR2 activity in the P. gingivalis LPS fraction is lipoprotein (14).…”

“…These findings suggested that evasion of innate immune response by P. gingivalis is due to gingipains-mediated inhibition of antimicrobial peptide expression. Recently, we reported that P. gingivalis stimulation up-regulates intracellular IL-33 expression in human gingival/oral epithelial cells via gingipain-dependent mechanisms (30). To determine whether or not intracellular IL-33 participates in P. gingivalis-induced LL-37 induction, we reduced IL-33 mRNA expression in Ca9-22 cells using an IL-33-specific siRNA.…”

“…In contrast, IL-33 is also involved in the development of chronic inflammatory diseases (26). We reported recently that increasing expression of IL-33 is detected in gingival epithelium from patients with chronic periodontitis, and that gingipains from P. gingivalis increased intracellular IL-33 expression in human gingival epithelial cells (30). The function of intracellular IL-33 induced by P. gingivalis in gingival epithelial cells may involve the modulation of innate immune function and the pathogenesis of periodontal diseases.…”
