Polyglycolic acid microneedles modified with inkjet-deposited antifungal coatings

In this study, the authors examined use of piezoelectric inkjet printing to apply an antifungal agent, voriconazole, to the surfaces of biodegradable polyglycolic acid microneedles. Polyglycolic acid microneedles with sharp tips (average tip radius = 25 ± 3 μm) were prepared using a combination of injection molding and drawing lithography. The elastic modulus (9.9 ± 0.3 GPa) and hardness (588.2 ± 33.8 MPa) values of the polyglycolic acid material were determined using nanoindentation and were found to be suitable for use in transdermal drug delivery devices. Voriconazole was deposited onto the polyglycolic acid microneedles by means of piezoelectric inkjet printing. It should be noted that voriconazole has poor solubility in water; however, it is readily soluble in many organic solvents. Optical imaging, scanning electron microscopy, energy dispersive x-ray spectrometry, and Fourier transform infrared spectroscopy were utilized to examine the microneedle geometries and inkjet-deposited surface coatings. Furthermore, an in vitro agar plating study was performed on the unmodified, vehicle-modified, and voriconazole-modified microneedles. Unlike the unmodified and vehicle-modified microneedles, the voriconazole-modified microneedles showed antifungal activity against Candida albicans. The unmodified, vehicle-modified, and voriconazole-modified microneedles did not show activity against Escherichia coli, Pseudomonas aeruginosa, or Staphylococcus aureus. The results indicate that piezoelectric inkjet printing may be useful for loading transdermal drug delivery devices such as polyglycolic acid microneedles with antifungal pharmacologic agents and other pharmacologic agents with poor solubility in aqueous solutions.

Electric field-driven fiber formation (electrospinning) is developing into a practical means for preparing novel porous filament with unusual structures and affordable mechanical properties. Polycaprolactone (PCL) was dissolved in solvent mixtures of methylene chloride/N,N-dimethyl formamide with ratios of 100/0, 75/25, and 50/50 (v/v) for electrospinning. The filament was formed by coagulation of the spinning solution following the well-known principle of phase separation in polymer solutions valid in other wet shaping processes. A strand of electrospun porous filament consisted of fibers ranging from 0.5 to 12 m in diameter. To evaluate the feasibility of three-dimensional fabric as scaffold matrices, the plain weave, which is the simplest of the weaves and the most common, was prepared with porous PCL filament. The growth characteristics of MCF-7 mammary carcinoma cells in the woven fabrics showed the important role of matrix microstructure in proliferation. This study has shown that woven fabrics, consisting of porous filaments via electrospinning, may be suitable candidates as tissue engineering scaffolds.

Preparation and In Vivo Study of Porous Titanium–Polyglycolide Composite

Porous materials show low Young s moduli and excellent bonding to living bone. However, the strength of such materials is often insufcient in the initial stage of implantation. Thus, the objective of this study was to increase the strength of porous titanium by lling the pores with polyglycolide (PGA), a biodegradable plastic. PGA powder was prepared via the thermal decomposition of sodium chloroacetate at 433 K. The PGA was then introduced into the pores of porous Ti (porosity: 60%) using two methods: (i) centrifugal packing and heating and (ii) heat injection. In the latter method, almost all pores were lled by PGA; the lling fraction was measured to be 65-85% regardless of the injection temperature. When the pores in the porous Ti were lled with PGA, the compressive strength increased drastically from 40 to 100 MPa. The increased strength is comparable to that of cortical bone. In addition, the strength increased with increasing injection temperature. In an animal test, unfavourable autopsy ndings, such as suppuration, bleeding, and hyperplasia of the connective tissue, could not be con rmed in rats and no bone was observed in the pores of the Ti-PGA composite. Decomposition of PGA lowered the surrounding pH, it was found to inhibit bone formation in the pores of the porous Ti. It is important to control the decomposition rate of PGA.
