Genetic structure and natal origins of green turtle mixed stocks in southern Brazil were assessed based on analyses of mtDNA control region sequences from the Arvoredo Island (n = 115) and Cassino Beach (n = 101) feeding areas. These were compared to other mixed aggregations to examine structuring, and to Atlantic Ocean nesting colonies to evaluate natal origins through Bayesian mixed stock analysis (MSA). In order to develop novel priors, surface drifter trajectories in the Atlantic were analyzed and combined with rookery data, and we used KulbackLeibler information measures in order to compare the difference of information among the 4 proposed priors. Each study area presented 12 haplotypes, 10 of which were shared at similar frequencies. Haplotypes CM-A8 and CM-A5 represented ~60 and 20%, respectively, and remaining haplotypes accounted for < 5% of samples. The 2 study areas were genetically similar to all feeding grounds in the western South Atlantic except Almofala, in northeast Brazil, and genetically different from Caribbean and North American mixed stocks. Drifter trajectory analysis revealed that drifters from Ascension and Trindade Islands have a larger chance of reaching Brazil. The priors drifter data and rookery size/drifter data combined contained the most information, but stock estimates were not greatly changed. MSA indicated that Ascension, Aves/Surinam and Trindade were the main stock contributors to the study areas. Advances in the Application of Genetics in Marine Turtle Biology and Conservation

Marine turtles migrate across long distances, exhibit complex life histories, and occupy habitats that are difficult to observe. These factors present substantial challenges to understanding fundamental aspects of their biology or assessing human impacts, many of which are important for the effective conservation of these threatened and endangered species. The early development and application of genetic tools made important contributions to understanding marine turtle population and evolutionary biology, such as providing evidence of regional natal homing by breeding adults, establishing connectivity between rookeries and foraging habitats, and determining phylogeography and broad scale stock structure for most marine turtle species. Recent innovations in molecular technologies, statistical methods, and creative application of genetic tools have significantly built upon this knowledge to address key questions in marine turtle biology and conservation management. Here, we evaluate the latest major advances and potential of marine turtle genetic applications, including improved resolution and large-scale syntheses of population structure, connectivity and phylogeography, estimation of key demographic rates such as age to maturity and operational or breeding sex ratios, insight into reproductive strategies and behavior, and assessment of differential human impacts among populations. We then discuss remaining challenges and emerging capabilities, such as rapid, multiplexed genotyping, and investigation of the genomic underpinnings of adaptive variation afforded by high-throughput sequencing technologies.

Although it is commonly assumed that female sea turtles always return to the beach they hatched, the pathways they use during the years preceding their first reproduction and their natal origins are most often unknown, as it is the case for juvenile green turtles found in Martinique waters in the Caribbean. Given the oceanic circulation of the Guiana current flowing toward Martinique and the presence of important nesting sites for this species in Suriname and French Guiana, we may assume that a large proportion of the juvenile green turtles found in Martinique are originating from the Suriname–French Guiana beaches. To confirm this hypothesis, we performed mixed stock analysis (MSA) on 40 green turtles sampled in Martinique Island and satellite tracked 31 juvenile green turtles tagged in Martinique to (a) assess their natal origin and (b) identify their destination. Our results from MSA confirm that these juveniles are descendant from females laying on several Caribbean and Atlantic beaches, mostly from Suriname and French Guiana, but also from more southern Brazilian beaches. These results were confirmed by the tracking data as the 10 turtles leaving Martinique headed across the Caribbean–Atlantic region in six different directions and 50% of these turtles reached the Brazilian foraging grounds used by the adult green turtles coming from French Guiana. One turtle left the French Guianan coast to perform the first transatlantic migration ever recorded in juvenile green turtles, swimming toward Guinea‐Bissau, which is the most important nesting site for green turtles along the African coast. The extensive movements of the migrant turtles evidenced the crossing of international waters and more than 25 exclusive economic zones, reinforcing the need for an international cooperative network to ensure the conservation of future breeders in this endangered species.
