Population pharmacokinetics of steady-state carbamazepine in Egyptian epilepsy patients

Abstract: The POP PK model we have developed for CBZ shows good predictive performance in Egyptian adult and pediatric patients with epilepsy. Another PK study to better define the effect of different covariates would improve on the model for dosage individualization.

“…Generally, DRE subjects received higher doses of CBZ than seizure free subjects and demonstrated an approximately 4% higher CL/F plasma as compared to their counterparts. Nevertheless, this difference was not significant, a result comparable to an Egyptian study . These observations support the fact that PWE with harder to control seizure does require significantly higher amount of CBZ in order to attain therapeutic concentrations, and it may be related to the lower free fraction of CBZ.…”

“…The population estimate of CL/F plasma of approximately 5.9 L/h concurred with those found in previous studies. As majority (72%) of this population was on polytherapy, CL/F plasma estimated are similar to the other polytherapy populations . Interestingly, CL/F dbs derived using similar PPK approach resulted in similar model specifications and identical parameter estimate with CL/F plasma .…”
