Population pharmacokinetics of 25-hydroxyvitamin D in healthy young adults

Abstract: The PPK model obtained determined clearance of 25-hydroxyvitamin D in a healthy young adult population in Serbia. Mean intake of vitamin D from foods and serum phosphate level are the most important covariates that influence value of 25-hydroxyvitamin D clearance in healthy young adults.

“…In our study 49.3% of the participants were smokers, 13.3% consumed more than 3 alcoholic drinks weekly and 29.3% more than 3 cups of coffee daily. However, our final PPK model did not recognize any of these three covariates as important elements in 25-hydroxy vitamin D clearance, an observation which is in accordance with the results of previous PPK analysis in a healthy, young population (10). The effects of caffeine consumption in amounts larger than 300mg/day on the serum level of 25-hydroxy vitamin D has been studied in a large number of epidemiological and experimental trials and the results provide a much clearer picture of this correlation.…”

“…The semi-structured questionnaire designed for assessment of vitamin D status was based on previous studies concerning the topic and conducted in the same geographical area (26,27). That original questionnaire was modified and supplemented for additional variables of interest, taking into account the following: a) the questionnaires for assessment of demographic, socio-economic, nutritional and life-style habits of the country's population, used by the national public health authority (28); b) the guidelines for the assessment of status, protective and risk factors for bone and vitamin D metabolism (29,30); and c) general methodological principles (31).…”

“…According to our knowledge, there were no studies published in recent years which investigated 25(OH)D serum concentrations in our country in order to reveal the current vitamin D status for healthy people in a large sample. Yet, some studies which included the postmenopausal women and the students, measuring 25(OH)D serum concentrations during sunshine months, also found a very high frequency of vitamin D deficiency in those, apparently healthy subjects (15,16). Nevertheless, we are unable to make any conclusion about vitamin D status on the population level at present as the matter was outside the scope of our research.…”

“…The semi-structured questionnaire for the assessment of vitamin D status was designed based on the previous studies in the field and the study was conducted within the same geographical area (7)(8). Other data were extracted from the patientsí medical files or were obtained through an interview in case of healthy subjects.…”
