Population Genetics and Forensic Efficiency of 30 InDel Markers in Four Chinese Ethnic Groups Residing in Sichuan

Abstract: Sichuan Province is located at the transitional junction regions of the Qinghai-Tibet Plateau and the low-altitude plains. It also serves as the corridor of Sino-Tibetan-speaking population migration and expansion since neolithic expansion of Proto-Tibeto-Burman populations from Middle/Upper Yellow River during Majiayao period (3300-2000 BC). However, the population structure and the corresponding genetic diversity of forensic-related markers in this region remain unclear. Thus, we genotyped 30 insertion-delet… Show more

“…To maximize the human identification effectiveness of InDel in Chinese population, novel InDels with higher heterozygosity levels should be identified to improve the marker set. In the present study, we select 34 autosomal InDel loci with the minimum allele frequency (MAF) ≥ 0.25 in East Asia, which show no overlap with the InDel markers from the Investigator ® DIPplex kit [ 34 , 35 ]. Based on the six-dye fluorescence labeling technology, a multiplex InDel kit containing all the above InDels is constructed and commercialized for Chinese population [ 36 ].…”

“…As the biallelic DNA length polymorphism in human genome, InDels possess highlighted merits such as wide distributions in human genome, low mutation rates, small amplified fragments, absence of stutter peaks in comparisons with STRs and available for capillary electrophoresis (CE) platform [ 5 , 6 ]. InDels complement the disadvantages of STRs, which have become the promising genetic markers in the applications of HID [ 7 , 8 ] and biogeographic ancestry predictions [ 9 ] since the initial InDel map was reported in 2006 [ 10 ].…”
