Population genetic study of four short tandem repeat loci in the Belgian population, using capillary electrophoresis

Abstract: Allele frequencies of four short tandem repeat loci (HumCD4, HumTH01, HumD21S11 and HumSE33) were investigated in a sample of 395 unrelated Belgian individuals using multiplex polymerase chain reaction and capillary electrophoresis. Automated laser fluorescence was used to detect four fluorescent dyes, enabling the use of an internal standard within each lane. With this method rapid typing with high resolution was obtained and the different alleles were grouped on a statistical base. All loci meet Hardy-Weinbe… Show more

“…In some criminal cases, however, objects and clothes are left at the scene of the crime with no obvious traces for DNA typing at first sight. In Belgium several cases of theft and burglary were solved by DNA typing of physical fingerprints and skin debris left on grips of bags, tools, and clothes [8]. This shows the great sensitivity of this technique and the possibility of using it on single physical fingerprints as first reported by van Oorschot and Jones in 1997 [9].…”

“…A multiplex of 4 STR loci (CD4, TH01, D21S11 and SE33) was used as described earlier [11] with slight modifications. In short, 1.3 units of Hotstar Taq DNA polymerase (Qiagen, Venlo, The Netherlands) was used and the samples were amplified on an Applied Biosystems GeneAmp 9700 60 Well thermal cycler.…”

“…For assessing the profile recovery after LPC from pure semen samples, a multiplex of four STR loci (CD4, TH01, D21S11, and SE33) was used as described earlier [13] with slight modifications. In short, 1.3 units of hotstar Taq DNA polymerase (Qiagen, Huntsville, AL, USA) was used and the samples were amplified on an Applied Biosystems GeneAmp 9700 60 Well thermal cycler (Foster City, CA, USA).…”
