“…Despite the microbiological methods used, plenty of the studies on microbial communities of activated sludge bioreactors have been conducted to profile taxonomic information based on 16S rRNA genes, using technical approaches of clone library (Wells et al 2011;Yang et al 2011), denaturing gradient gel electrophoresis (DGGE) (Yang et al 2011), terminal restriction fragment length polymorphism (T-RELP) (Wells et al 2011;Yang et al 2011), fluorescence in situ hybridization (FISH) (Mielczarek et al 2012(Mielczarek et al , 2013, Illumina (Albertsen et al 2012), 454-pyrosequencing (Hu et al 2012;Wang et al 2012a), and PhyloChips (Xia et al 2010). However, 16S rRNA gene-based ways may not be efficient in distinguishing some closely related but ecologically distinct groups of microorganisms as a consequence of the low evolutionary rate of 16S rRNA genes (Palys et al 1997).…”

“…Earlier studies using the methods of T-RFLP and clone library indicated that the detected richness (numbers of operational taxonomic units (OTUs)) and dominant phyla and OTUs did not change much (Wells et al 2011) and revealed relatively stable AOB structures in full-scale wastewater treatment systems with relatively stable treatment efficiencies during a year of monitoring. Other studies applying FISH suggested that the relative abundances of filamentous bacteria (Mielczarek et al 2012), and abundant polyphosphate-accumulating organisms (PAOs) (Mielczarek et al 2013) showed minor changes in full-scale WWTPs with good treatment efficiencies during 3-year investigations. From this perspective, we considered that a single grab of sample from functionally stable systems could be effective, as it could at least reveal microbial richness and abundant groups.…”

“…Badania przeprowadzone z pomocą nowoczesnych technik biologii molekularnej w 28 oczyszczalniach ścieków w Danii wykazały, że bakterie nitkowate stanowią przeciętnie około 24% bakterii tworzących kłaczki osadu czynnego [11]. Wiele wskazuje na to, że bakterie nitkowate odgrywają kluczową rolę w formowaniu kłaczków osadu czynnego tworząc szkielet, do którego przyłączają się bakterie zooglealne [2].…”

“…The relatively high observed abundances of these groups indicates that they are likely the most important filamentous organisms in these systems (Mielczarek et al, 2012;McIlroy et al, 2015b) and they simply must make a substantial contribution to the bulk nutrient transformations observed. Although the ecophysiology of the 'Candidatus Microthrix parvicella' and P2CN44 phylotypes has been described (Kragelund et al, 2011;McIlroy et al, 2013), minimal ecological and physiological information is available for members of the B45 phylotype.…”

“…Semiquantitative survey data suggest that these organisms are particularly well suited to flourish under the dynamic electron acceptor conditions, where carbon substrates prevail under the anaerobic conditions, characteristic of plants designed to remove phosphorus (P) and nitrogen (N) (Speirs et al, 2009;Mielczarek et al, 2012). This suggests an ability for anaerobic carbon storage and/or respiration/fermentation.…”
