Polysulfides are possible H 2 S‐derived signaling molecules in rat brain

Abstract: Accumulating evidence shows that hydrogen sulfide (H2S) has a variety of physiological functions. H2S is produced from cysteine by 3 sulfurtransferases. H2S, in turn, generates polysulfides, the functions of which are not well understood. H2S induces Ca(2+) influx in astrocytes, a type of glia. However, the receptor that mediates the response has not been identified. Here, we have shown that polysulfides induce Ca(2+) influx by activating transient receptor potential (TRP)A1 channels in rat astrocytes (EC50 91… Show more

“…Effect of sulfide 1, 2 and 5 μM versus 0.5 μM the 'SSNO -mix' was determined using the Dunnett multiple comparison; λλ P < 0.01. must account for the relaxation observed. These may include Sn and SSNO - [12,13,50]. Using EPR spectroscopy we confirmed the release of NO from the reaction mixture, but SSNO -is not known to release HNO.…”

“…We recently reported that polysulfides induced Ca 2+ influx by activating transient receptor potential (TRP) A1 channels, which have sensitive thiol residues, in rat astrocytes, and a maximal response was induced at 0.5 lM, which is 1/320 of the concentration of H 2 S required to achieve a response of similar magnitude [24]. These findings support the notion that bound sulfur species, such as polysulfides, have direct reactivity independent of H 2 S. Furthermore, a recent report determined that evidence that has been attributed to H 2 S in many previous reports may in fact have been mediated by polysulfides [25].…”

“…Accumulation of this product was also observed after reaction of RSNOs with sulfide at pH 7.4 (26,27); moreover, SSNO − -containing mixtures were found to release NO, activate soluble guanylyl cyclase (sGC) (26), and relax vascular tissue (28), although a contribution of other reaction products to these effects cannot be excluded. Meanwhile, other sulfane sulfur molecules, including persulfides (RSSH) and polysulfides (RSS n − and HS n − ), have come to the fore as potential mediators of sulfide's biological effects (29)(30)(31), but little is known about their pathways of formation, prevalence in biological systems, and relationship with NO.…”
