Polysome Profiling Shows the Identity of Human Adipose-Derived Stromal/Stem Cells in Detail and Clearly Distinguishes Them from Dermal Fibroblasts

Abstract: Although fibroblasts and multipotent stromal/stem cells, including adipose-derived stromal cells (ADSCs), have been extensively studied, they cannot be clearly distinguished from each other. We, therefore, investigated the cellular and molecular characteristics of ADSCs and fibroblasts. ADSCs and fibroblasts share several morphological similarities and surface markers, but were clearly found to be different types of cells. Contrary to previous reports, fibroblasts were not able to differentiate into adipocytes… Show more

“…Mechanisms for fatty metaplasia have been proposed including cell damage with intracytoplasmic lipid accumulation and differentiation from facultative fibroblasts. More recent stem cell investigations have identified dermal stem cells distinctive from dermal fibroblasts that may be stimulated to differentiate to adipocytes …”

“…After induction of osteogenic differentiation for 24 h, cells were trypsinized, plated on 24-well plates (2,5 × 10 4 cells/well) and kept for 10, 20 and 40 min in a humidified incubator at 37 °C with 5% CO 2 as previously described 56 . The non-adherent cells were removed by washing twice with PBS.…”

“…A classical technique called polysome profiling has been used to assess the mRNAs bound to ribosomes through ultracentrifugation in a sucrose gradient, and further identify them by high-throughput methods [ 23 – 25 ]. Previously, we used polysome profiling to study the fate of human adipose stem cells (hASCs) and their commitment to adipogenesis [ 25 , 26 ]. We showed that 60% of the genes, which were differentially expressed after 72 h of differentiation induction, were controlled by post-transcriptional regulation.…”
