Polysaccharide isolated from Korean-style soy sauce activates macrophages via the MAPK and NK-κB pathways

Abstract: Soy sauce is a staple fermented food in Korea, however, there are relatively limited studies on soy sauce in Korea compared to that in Japan. In this study, the immuno-stimulating activities are compared in polysaccharides isolated from soy sauce in Korea and Japan. Crude polysaccharides was isolated, and which were named as Korean-style soy sauce (KSS-0) and Japanese-style soy sauce (JSS-0). Then, difference in cytokine production was compared in peritoneal macrophages, and KSS-0 showed higher cytokine produc… Show more

“…Nuclear factor-kappa B (NK-κB) plays a vital role in the process of inflammatory response (Shin et al, 2021). Furthermore, some reports have verified that hindered NF-kB signaling pathway reduced renal injury in IR-mediated AKI (Zeng et al, 2020).…”

“…Polysaccharides derived from legumes exhibit various immunomodulatory functions (62). Particularly, soy polysaccharides induce the production of pro-inflammatory cytokines (IL-1β, IL-6, IL-12, and TNF-α) and mediators (NO and ROS) in unstimulated RAW264.7 cells (50,51). Notably, the immunostimulatory effects are dependent on JUN/ERK/p38-mediated activation of NF-kB signaling (51) (Table 1).…”

“…MAPK is one of the vital signal transduction systems in organisms, and it participates in mediating various cellular processes such as growth, development, division, differentiation, death, and intercellular functional synchronization (Yang et al, 2017;Zhao et al, 2019). The threonine and tyrosine on the T-loop structure of the three subfamilies of MAPK (p38, JNK, ERK) are activated by dual phosphorylation under the stimulation of TNF-α (Foletta et al, 1998;Shin et al, 2021). Then, the MAPK subfamily undergoes nuclear enters the nucleus to link the inflammatory response element on DNA, upregulating the expression of downstream inflammation genes (such as TNF-α, IL-1, IL-6, and other adipokines) (Ono & Han, 2000).…”
