We previously found that passion fruit (Passiflora edulis) seeds contained a high amount of piceatannol (3,5,3′,4′-trans-tetrahydroxystilbene), a natural analog of resveratrol (3,5,4′-trans-trihydroxystilbene). Resveratrol has been proposed as a potential anti-metabolic disorder compound, by its activation of sirtuin and AMP-activated protein kinase. Many reports show that resveratrol ameliorates diet-induced obesity and insulin resistance. However, it is not known whether piceatannol also affects diet-induced obesity. We explored the effect of piceatannol on high fat diet-fed mice. The results showed that piceatannol did not affect high fat diet-induced body weight gain or visceral fat gain in mice. However, piceatannol did reduce fasting blood glucose levels. Furthermore, to explore the potential of passion fruit seed extract containing piceatannol as a functional food, passion fruit seed extract was administered in a genetic diabetic mouse model (db/db mice). Single administration of passion fruit seed extract, as well as piceatannol reduced the blood glucose levels of these db/db mice. These results suggest that piceatannol and passion fruit seed extract may have potential application in the prevention of diabetes.Key words piceatannol; passion fruit seed; antidiabetic activity; high fat diet; db/dbThe incidence of metabolic syndrome is increasing worldwide due to aging and changes in lifestyle. In particular, this increase is largely attributed to the rise in the prevalence of obesity, and this common metabolic disorder is associated with increased risk of cardiovascular diseases and type II diabetes, and the latter substantially decreases quality of life and increases health care costs. It is difficult to strictly control blood glucose levels over the long term, even though many anti-diabetic medications are clinically available. Therefore, the development of functional foods that can prevent or improve type II diabetes is an urgent matter.Resveratrol is a natural component of several plants, including grapes, berries, and peanuts. Schwartzia brasiliensis (Marcgraviaceae) is a native Brazilian shrub species with neotropical distribution in shoal environments. Recent studies have revealed its medicinal potential for some human diseases; therefore, this study aimed to evaluate the genotoxic and antigenotoxic potential of extracts and fractions obtained from different organs of S. brasiliensis. To accomplish this, plant extracts at different concentrations (25, 125 and 250 µg/ml) were incubated with plasmid DNA, either alone, or in the presence of stannous chloride as a positive control. Samples were then examined to detect any plasmid strand breaks or the absence of such breaks, which would indicate protection of these molecules against stannous chloride-induced lesions. Methanol and aqueous extracts of leaves and stems showed the ability to withstand the effects of stannous chloride in that no DNA damage was observed. Moreover, no other extracts or fractions used in the experimental conditions assayed resulted in DNA damage. These findings suggest that S. brasiliensis has antigenotoxic properties, indicating, in turn, that its biological activities deserve further study given the medicinal relevance of this plant.

Type II Diabetes Mellitus (T2DM), is a growing global epidemic (1,2) , exacerbated by the existing obesogenic environment and physical inactivity. There is a need for improved lifestyle interventions and new dietary strategies to prevent and treat T2DM. Evidence suggests that natural bioactives may play a role in the management of blood glucose levels, by numerous mechanisms, including their insulinotropic effects (3)(4)(5) . This aim of this research was to elucidate the potential glycaemic management properties of a range of plant derived bioactives.In vitro dose response experiments were conducted in the BRIN-BD11 cell line. Cells were cultured in T175 flasks in RPMI-1640 media with 10 % (v/v) foetal calf serum, 2 mM L-glutamine, 50 IU/ml penicillin, 0·05 mg/ml streptomycin, and incubated at 37°C and 5 % C0 2 . Acute insulin secretion experiments (n = 4) were performed. Cells were incubated for 20 mins in the presence of a range of concentrations of plant bioactives including epicatechin, epigallocatechin gallate (EGCG), quercetin and genistein in 16·7 mM glucose. (Sigma-Aldrich Ireland Limited, Wicklow, Ireland). Insulin release was measured by ELISA assay (Mercodia Ultrasensitive Rat Insulin, Sweden). Statistical analysis was performed using SPSS V20 software.Quercetin and genistein stimulated insulin secretion in a dose response manner, while EGCG and epicatechin did not. Exposure to quercetin [100uM] induced an insulin release of 47·76 ± 3·45 ng/mg protein which was significantly higher than the control, 16·7 mM glucose, with an insulin release of 24·31 ± 1·96 ng/mg protein, p < 0·05. Exposure to a combination of quercetin [100uM] and genistein [20uM] demonstrated a synergistic effect on insulin release of 65·892 ± 8·75 ng/mg protein; which was significantly higher, p < 0·05, than quercetin [100uM] alone.The potential of quercetin and genisteins' synergistic effect should be investigated in vivo and the cellular mechanisms are currently being investigated. This research will lay the foundations for the development of innovative foods, which will be targeted at prediabetics and people with T2DM.
