AbstractBackgroundRadiation-induced injuries (RII) mainly result from reactive oxygen species (ROS), which are harmful compounds that can damage DNA. Asiaticoside (AC), one of the main functional components extracted from Centella asiatica, has potent pharmacological effects such as anti-inflammatory and anti-oxidant activity. However, its role in RII remains unclear.PurposeThe purpose of the current study is to investigate whether AC can mitigate RII in vitro and in vivo.Material and MethodsCell model of RII was successfully established by 5J/m2 radiation in vitro. For the in vivo RII model, mice were irradiated with 5 Gy to the thorax. The degree of damage to cells or mouse tissue was determined by measuring the numbers of DNA double-strand breaks (DSBs), oxidative stress, and mouse survival rates.ResultsIn the in vitro assay, AC administration significantly reduced radiation-induced growth inhibition of Escherichia coli and fibroblasts, DSBs and apoptosis of fibroblasts; in the in vivo study, AC could decrease antioxidant capacity (T-AOC) of plasma and protect mice from RII, thereby improving the survival rates of mice after radiation.ConclusionsThese novel data indicate that AC is able to prevent radiation-initiated genotoxicity by mitigating DNA damage, and might serve as a safe and effective radio-protective agent.

Berberine (BER), a natural isoquinoline alkaloid, has been demonstrated to have appreciable anticolitis effects. Nevertheless, the protective mechanism of BER in ulcerative colitis (UC) is barely understood. The present study was aimed at exploring the therapeutic efficacy of BER on UC in experimental colitis rat model. Rats were orally administered with BER for seven days at low and high doses (25 and 50 mg/kg/day) before AcOH intracolonic instillation. BER significantly retrieved colon inflammation and mucosal damage indicated by inhibition of macroscopic score and lessened the levels of inflammatory biomarkers (IL-1β, IL-6, TNF-α, MPO, and PGE2). Notable downregulation of mRNA expression of p38 MAPK and increased protein expression of TGF-β were achieved by BER treatment. The anti-inflammatory potential of BER was supported by the histopathological screening of colon mucosa. In addition, BER restored colonic antioxidant capacity through elevation of GSH level and antioxidant enzymatic activities (SOD, CAT, GPx, and GR) together with reductions of both MDA and NO levels. Marked downregulation of Nos2 mRNA expression is accompanied by increased Nrf2 and Hmox-1 expressions in colon specimens treated by BER. Furthermore, BER exhibited noticeable antiapoptotic activities through decreasing proapoptotic proteins (Bax and caspase-3) and lessening antiapoptotic Bcl-2 protein in the colon mucosa. Based on these findings, BER may improve colitis markedly which may be mediated by its striking antioxidant, anti-inflammatory, and antiapoptotic properties.

The beneficial effects of Ocimum gratissimum, have been attributed mainly to its antioxidant and antiinflammatory properties. This study investigated the protective effects of aqueous extract of Ocimum gratissimum leaves (AEOGL) on acetic acid induced colitis in male rats. Twenty male Wistar rats, 100-180 g were divided into four groups as follows: Group 1 (control) (n = 5) received 2 ml/kg of distilled water for 21 consecutive days. Group 2 (n = 5) received 2 ml of 6% acetic acid solution once intra rectally for induction of colitis. Group 3 and 4 (n = 5 each) were treated as group 2 and thereafter received AEOGL orally at 200 and 400 mg/kg/day respectively for 20 consecutive days. All the animals from each group were sacrificed 24 h after the induction of colitis and administration of AEOGL. The diarrhea score, ulcer score, hematological parameters, nitric oxide (NO), myeloperoxidase (MPO), superoxide dismutase (SOD), reduced glutathione (GSH) markers and histopathological alteration were evaluated. Acetic acid-induced colitis significantly caused alteration in diarrhea score, ulcer score, hematological parameters, MPO and SOD activities, NO and GSH levels (p < 0.05). It induced significant inflammation of the colonic tissue. AEOGL administration significantly decreased diarrhea score and ulcer score to normal (p < 0.05). It prevented alteration effect of acetic acid on hematological parameters and significant decreases the activities of MPO, SOD, NO and increase GSH levels (p < 0.05) in colitis rats. In conclusion, Ocimum gratissimum leaves possesses ameliorative effects against acetic acid-induced colitis as a consequence of its antiinflammatory and anti-oxidative properties.
