Polymorphonuclear Leukocyte Behavior in a Nonhuman Primate Focal Ischemia Model

Abstract: Summary: There is increasing interest in the role of poly morphonuclear (PMN) leukocytes in the evolution of fo cal cerebral infarction. Surgical preparation of focal ce rebral ischemia models may alter leukocyte reactivity and thereby make interpretation of leukocyte function follow ing ischemiaireperfusion difficult. The effects of surgical preparation and of experimental ischemiaireperfusion on granulocyte function have been examined prospectively in a baboon model. Twenty-six adolescent male baboons underw… Show more

“…A causative involvement of MMP-9 in these events remains to be tested. (Hibbs et aI., 1985;Kjeldsen et aI., 1992;Vartio and Baumann, 1989) and increase early during MCAO in this model (Ember et a!., 1994), no clear correlation between MMP-9 activity and leukocyte count could be seen here.…”

“…11 In stroke models, additional factors such as exposure to anesthetics and surgery may impair leukocyte functions and, thus, may contribute to the increased risk of infection. 18 However, in contrast to MCAO mice, shamoperated animals neither showed a relevant attenuation of immune responsiveness nor developed spontaneous bacterial infections 11 indicating that the surgical procedures and anesthetics do not play a major role in the impaired antibacterial defenses after stroke in this model.…”

“…The possibility that leukocyte alterations in experimental models may be determined by the surgical procedures used to induce the ischemic insult or by anesthesia has been refuted in a nonhuman primate model. 153 Moreover, the use of shamoperated animals (ie, animals in which all of the phases of the surgical procedure except artery occlusion are reproduced) makes it possible to control for the influence of surgery and anesthesia. Some of the substantial advantages of experimental models are the following: (1) The possibility of knowing the exact time of onset of ischemia means that the temporal pattern of the expression of the molecules under study can be investigated; (2) Access to brain tissue for pathological observation is simple; (3) New drugs can be tested.…”
