Polymorphisms of TLR4 but not CD14 are associated with a decreased risk of aggressive periodontitis

Abstract: It is concluded that in West European Caucasians, the Asp299Gly TLR4 gene polymorphism is associated with a decreased risk of AgP but not CP. Promoter polymorphisms of the CD14 gene, however, did not influence susceptibility to inflammatory periodontitis in the population cohorts studied.

“…Significantly higher prevalence of this allele was shown in chronic periodontitis (p = 0.0495) and the absence of significant results for people with a general diagnosis of periodontitis (p = 0.156) and for the AgP group (p = 0.755) was reported. Only two homozygous subjects were found, one in the CP [22] and one in the control group [21].…”

“…The rest of the publications included 2 polymorphisms. In 8 articles [18, 21, 23, 25-27, 30, 31] study groups consisted of subjects with chronic periodontitis, in 2 [24,29] aggressive periodontitis and in 5 [19,20,22,28,32] The frequency of Asp299Gly polymorphism was 12.83% among subjects with periodontitis (13.76% in CP and 10.57% in AgP) and 12.14% in the control group. The frequency of Asp299Gly allele in periodontitis was 6.44% (6.93% in CP and 5.28% in AgP) and 5.47% in the control group.…”

“…The role of each TLR in innate immunity against bacteria and how TLRs active inflammatory cytokines and antimicrobial peptides have been well documented [2,3]. In addition, Asp299Gly and Thr399Ile mis-sense mutations in extracellular domain of TLR4 have been associated with decreased late diabetic complication and aggressive periodontitis [4,5].…”
