Polymorphisms in theSP110andTNF-αGene and Susceptibility to Pulmonary and Spinal Tuberculosis among Southern Chinese Population

Abstract: Objective To investigate the association of single-nucleotide polymorphisms (SNPs) in SP110 gene and TNF-α gene among pulmonary TB (PTB) and spinal TB (STB) patients. Methods In a total of 190 PTB patients, 183 STB patients were enrolled as the case group and 362 healthy individuals at the same geographical region as the control group. The SP110 SNPs (rs722555 and rs1135791) and the promoter -308G>A (rs1800629) and -238G>A (rs361525) polymorphisms in TNF-α were genotyped. Results. TNF-α -238G>A polymorphism wa… Show more

“…Thus, loss of Ipr1/Sp110 may account for the Sst1 S phenotype, though this remains to be confirmed by the generation of B6.Ipr1 −/− mice. Polymorphisms in human SP110 are associated with TB disease in some but not all cohorts [61][62][63][64][65][66][67][68] . Ipr1/Sp110 is a member of the Sp100 family of transcriptional regulators that also includes Sp100, Sp140, and Aire.…”

“…Meanwhile, the study of Zhou et al (2017) provides further evidence supporting the host genetic variability in TB susceptibility, especially in different types of TB infection which is the TNF-α -238 A allele indicates a protective effect against STB, but not against PTB, whereas the SP110 SNPs (rs722555 and rs1135791) and TNF-α -308G>A (rs1800629) showed no association with susceptibility to PTB and STB in southern China. In the same way, there was a susceptible genetic factor and the possible outcome of PTB infection.…”

“…In recent years, drug-resistant tuberculosis, particularly multidrug-resistant tuberculosis, has emerged globally ( 5 ). Additionally, an increase in longer chemotherapy cycles and drug side effects have presented major challenges to effectively control osteoarticular tuberculosis ( 6 , 7 ). Therefore, the pathogenic mechanism of osteoarticular tuberculosis must be determined to develop new and effective prevention and treatment strategies.…”
