Polymorphisms in dipeptidyl peptidase 4 reduce host cell entry of Middle East respiratory syndrome coronavirus

Abstract: Middle East respiratory syndrome (MERS) coronavirus (MERS-CoV) causes a severe respiratory disease in humans. The MERS-CoV spike (S) glycoprotein mediates viral entry into target cells. For this, MERS-CoV S engages the host cell protein dipeptidyl peptidase 4 (DPP4, CD26) and the interface between MERS-CoV S and DPP4 has been resolved on the atomic level. Here, we asked whether naturally-occurring polymorphisms in DPP4, that alter amino acid residues required for MERS-CoV S binding, influence cellular entry of… Show more

“…The most important of these is a glutamine at position 493 at the receptor binding domain, which explains its increased transmissibility compared to SARS CoV [27]. b) Changes in ACE-2 Receptor: With regard to MERS-CoV, which employs CD26 (DPP-4) as its receptor for cellular entry, there are naturally-occurring polymorphisms in DPP4 that impact cellular entry of MERS-CoV and might thus modulate MERS development in infected patients [28]. There is a possibility that similar variations or polymorphisms in ACE-2 could affect the viral entry and disease course.…”

“…In this way, apart from pre-existing health conditions potentially predisposing to SARS-CoV-2 (e.g. obesity, hypertension, chronic respiratory disease, compromised immune status), we believe that there might be some protecting or predisposing genetic variants in genes such as ACE2, TMPRSS2 and DPP4, recently described as potentially modulating COVID-19 illness [7][8][9][10].…”

“…After attachment of MRES-CoV spike (S) glycoprotein to DPP4 positive human cells the S protein undergoes proteolytic activation through the cellular serine protease TMPRSS2 and cysteine protease cathepsin L once inside endosomes 60 . It was recently reported that among fourteen characterized mutants forms of DPP4, four polymorphisms (K267E, K267N, A291P and D346-348) strongly reduce the binding and penetration of MERS-CoV into target cells and the viral replication 61 .…”
