PolyMorphine: An innovative biodegradable polymer drug for extended pain relief

Abstract: Morphine, a potent narcotic analgesic used for the treatment of acute and chronic pain, was chemically incorporated into a poly(anhydride-ester) backbone. The polymer termed “PolyMorphine”, was designed to degrade hydrolytically releasing morphine in a controlled manner to ultimately provide analgesia for an extended time period. PolyMorphine was synthesized via melt-condensation polymerization and its structure was characterized using proton and carbon nuclear magnetic resonance spectroscopies, and infrared s… Show more

“…[3] Recently, living radical and ring-opening metathesis polymerization of prodrugs have been explored to prepare polymer therapeutics, [4] however, polymer-drug conjugates synthesized by these methods are non-biodegradable, which limits their clinical application. We chose organocatalytic ROP [5] for synthesis of polymer prodrugs because it is a powerful method for the synthesis of aliphatic polyesters, [6] polycarbonates, [7] polypeptides, [8] and polyphospho-esters, [9] that are biodegradable.…”

“…To overcome these limitations, direct polymerization of polymerizable drugs has been pursued. For example, Uhrich and co-workers used the conventional condensation polymerization to create biodegradable polyprodrugs [142144]. However, only a very limited number of drugs can be directly polymerized, and the polymerized products typically do not carry assembling potential, and may still need delivery vehicles to maximize their efficacy.…”
