Polymorphic microsatellite DNA markers for the Florida manatee (Trichechus manatus latirostris)

Abstract: Florida manatees ( Trichechus manatus latirostris ) are marine mammals that inhabit the coastal waters and rivers of the southeastern USA, primarily Florida. Previous studies have shown that Florida manatees have low mitochondrial DNA variability, suggesting that nuclear DNA loci are necessary for discriminatory analyses. Here we report 10 polymorphic microsatellite loci with an average of 4.2 alleles per locus, and average heterozygosity of 50.1%. These loci have been developed for use in population studies, … Show more

“…Amplifications were performed in a total volume of 15 lL, with 10 ng target DNA, 19 Sigma PCR Buffer (10 mM TrisHCl, pH 8. Pause et al 2007). For fragment analysis, the forward primers were labeled with the fluorescent dyes HEX or 6-FAM for processing and visualization on an ABI 3730xl Automated DNA Analyzer.…”

“…Fragment analysis of 15 polymorphic microsatellite loci ( TmaKB60, TmaA02, TmaE01, TmaE02, TmaE07, TmaE08, TmaE11, TmaE26, TmaH13, TmaJ02, TmaK01, TmaF14, TmaM79, TmaSC5, TmaSC13 ) [16], [17], was achieved following Kellogg [28] PCR conditions, and visualized on an Applied Biosystems ABI 3100 Genetic Analyzer (Universidad de los Andes). All individuals were genotyped for at least 12 loci.…”

“…Using multiple unlinked loci for populations with smaller sampling sizes improves accuracy in population genetics estimates [13], [14], [15], and provides a solution to study population dynamics in manatee populations. Primers for amplification of highly polymorphic bi-parentally inherited nuclear microsatellite loci have been developed for the Florida manatee [16], [17], [18], and effectively used to study manatee population structure in Florida, Puerto Rico, Belize and Mexico [19], [20], [21].…”

“…A panel of 16 polymorphic microsatellite primers (Garc´ıa-Rodr´ıguez et al, 2000;Pause et al, 2007) was PCR amplified using 118 individuals. The study included 88 individuals from the SLS system (12 Northern Lagoon and 76 Southern Lagoon), 21 from BCC, two from PL and seven amplifiable carcass samples.…”
