We describe the synthesis of water-soluble molecularly imprinted polymer nanoparticles (MIP-NPs) as a new artificial host receptor for the recognition of adenosine monophosphate (AMP), used herein, as a model nucleotide. MIP-NPs were prepared by solid-phase synthesis on glass beads (GB) using, for the first time, immobilized Fe(III)chelate as an affinity ligand to orientate the AMP via its phosphate group. A polymerizable thymine monomer which can induce complementary base-pairing with the adenine moiety of the nucleotide was synthesized and incorporated in the polymerization mixture to constrain the AMP in a dualorientated configuration. The MIP-NPs were remarkably selective toward AMP as they did not bind other nucleotides, GMP, UMP, and CMP. This strategy of using the phosphate group of AMP as a hinge enables unhindered pairing of the nucleobase with its corresponding complementary base monomer and can be extended to the preparation of specific MIP receptors for other key nucleotides in aqueous conditions.

In this study, we have developed a method to assess adenosine 5ʹ‐triphosphate by adsorptive extraction using surface adenosine 5′‐triphosphate‐imprinted polymer over polystyrene nanoparticles (412 ± 16 nm) for selective recognition/separation from urine. Molecularly imprinted polymer was synthesized by emulsion copolymerization reaction using adenosine 5′‐triphosphate as a template, functional monomers (methacrylic acid, N‐isopropyl acrylamide, and dimethylamino ethylmethacrylate) and a crosslinker, methylenebisacrylamide. The binding capacities of imprinted and non‐imprinted polymers were measured using high‐performance liquid chromatography with UV detection with a detection limit of 1.6 ± 0.02 µM of adenosine 5′‐triphosphate in the urine. High binding affinity (QMIP, 42.65 µmol/g), and high selectivity and specificity to adenosine 5′‐triphosphate compared to other competitive nucleotides including adenosine 5ʹ‐diphosphate, adenosine 5ʹ‐monophosphate, and analogs such as adenosine, adenine, uridine, uric acid, and creatinine were observed. The imprinting efficiency of imprinted polymer is 2.11 for urine (QMIP, 100.3 µmol/g) and 2.51 for synthetic urine (QMIP, 48.5 µmol/g). The extraction protocol was successfully applied to the direct extraction of adenosine 5′‐triphosphate from spiked human urine indicating that this synthesized molecularly imprinted polymer allowed adenosine 5′‐triphosphate to be preconcentrated while simultaneously interfering compounds were removed from the matrix. These submicron imprinted polymers over nano polystyrene spheres have a potential in the pharmaceutical industries and clinical analysis applications.

Modified adenosine nucleosides have been proposed to be potential DNA-based biomarkers for early diagnosis of tumor and a promising tool for the development of noninvasive prediction systems. However, the low concentration of modified adenosine nucleosides in physiological fluids makes them challenging for both quantitative and qualitative determination. Therefore, materials, which are potentially useful for selective adsorption of nucleobase-containing compounds, were obtained. To obtain the adsorbents, the silica gel particles were coated layer-by-layer with films of the polymers with different combinations of polymers containing thymine groups. Next, the microspheres were irradiated with UV light in the presence of 2'-deoxyadenosine or 5'-deoxy-5'-(methylthio)adenosine, as template molecules, which resulted in the photodimerization of thymine moieties and molecular imprinting of adsorbed modified adenosine compounds. The selectivity of the adsorption was significantly enhanced by the photoimprinting process. Eventually, the imprinted particles have shown an improved ability to recognize mainly 2'-deoxyadenosine and 5'-deoxy-5'-(methylthio)adenosine molecules. The best performing adsorbent was obtained using modified natural polysaccharides. The studied materials could serve as promising adsorbents of biomarkers for tumor diagnostics.
