Polydatin Induces Apoptosis and Inhibits Growth of Acute Monocytic Leukemia Cells

Abstract: Polydatin (PD), a component isolated from Polygonum cuspidatum, has various activities such as inhibiting platelet aggregation, lowering level of blood lipid, reducing lipid peroxidation, and so on. However, the antitumor activity of PD has been poorly reported. In the present study, effect of PD on cell proliferation was evaluated by Cell Counting Kit-8, and cell cycle and apoptosis were investigated by flow cytometry. Meanwhile, the protein expression level of Bc1-2, Bax, cyclin A, cyclin B, and cyclin D1, w… Show more

“…Moreover, polydatin is a natural precursor of resveratrol, which is present in many vegetable extracts (e.g., vineyard products, Polygonum cuspidatum extracts, etc. ), with possible important beneficial effects on human health, namely the protection against hemorrhagic shock, ischemia/reperfusion, heart failure, endometriosis and cancer (e.g., see [50] and references therein).…”

“…Polydatin can inhibit cell proliferation via attenuating the β-catenin signaling and promotes cell apoptosis via up-regulation the ratio of Bax/Bcl-2 in osteosarcoma ( 14 ). Polydatin also induces apoptosis and inhibits growth of acute monocytic leukemia cells ( 12 ). Polydatin was found to exhibit anti-growth activity against 3D cell aggregates of the SKOV-3 and OVCAR-8 ovarian cancer cell lines ( 17 ).…”

“…The induction of apoptosis is suggested to be a good strategy in cancer treatment ( 20 ). Importantly, polydatin effectively induced cell apoptosis in various cancer cell lines including osteosarcoma cells, acute monocytic leukemia cells, lung cancer cells and colorectal cancer cells ( 12 – 14 , 18 ). In the present study, the data consistently showed that polydatin induced cell apoptosis in a concentration-dependent manner.…”

“…In recent years, studies on PD have revealed that it plays important roles in a wide range of cellular activities, such as anti-thrombotic formation and anti-neoplastic, strengthening the systolic and diastolic function of cardiac myocytes, improving microcirculation of important organs postshock. [37][38][39][40][41][42][43][44][45] Indeed, our drug-target-pathway network based on the targetprediction and GO/KEGG pathway analysis showed that PD possesses multiple protein targets that involve in an array of interactive pathways. Thus, we have reasons to believe that PD is associated with a variety of pharmacological activities against many diseases.…”
