Polychlorinated biphenyls (PCBs) are a family of toxicants that persist in measurable quantities in human and wildlife tissues, despite their ban in production in 1977. Some PCB mixtures can act as endocrine disrupting chemicals (EDCs) by mimicking or antagonizing the actions of hormones in the brain and periphery. When exposure to hormonally active substances such as PCBs occurs during vulnerable developmental periods, particularly prenatally or in early postnatal life, they can disrupt sex-specific patterning of the brain, inducing permanent changes that can later be manifested as improper sexual behaviors. Here, we investigated the effects of prenatal exposure to the PCB mixture Aroclor (A) 1221 on adult female reproductive behaviors in a dose-response model in the SpragueDawley rat. Using a paced mating paradigm that permits the female to set the timing of mating and control contact with the male during copulation, we were able to uncover significant differences in female-typical sexual activities in A1221-exposed females. Specifically, A1221 causes significant effects on mating trial pacing, vocalizations, ambulation and the female's likelihood to mate. The results further demonstrate that the intermediate treatment group has the greatest number of disrupted endpoints, suggestive of non-linear dose responses to A1221. These data demonstrate that the behavioral phenotype in adulthood is disrupted by low, ecologically relevant exposures to PCBs, and the results have implications for reproductive success and health in wildlife and women. Banned in 1977 in response to dawning public awareness of their estrogenic and potentially toxic effects on humans and wildlife, PCBs continue to leach into soil, air and groundwater via retired industrial equipment, and from old factories and buildings. PCBs may have variable degrees of impact depending on which congeners or congener mixtures are involved, the organism's age at exposure, the sex of the individual, the degree of exposure and the availability of compensatory diet or social buffering to counteract those effects. An accurate evaluation of ecologically relevant xenobiotic exposure (Battershill, 1994;Brouwer et al., 1999).The neuroendocrine system serves as an interface between the central nervous system and peripheral endocrine organs, and thus represents a prime target for endocrine disruption by PCBs (Patisaul et al., 2006). In 1979, a nun poisoning involving 2,000 people occrred in central Taiwan from ingestion of cooking oil contaminated by polyckorinated biphenyls (PCBs) and polyorinatd dibenaofurans (PCDFs). We studied the prevdence of medic conditions in the exposed indivduals and in a neighborhood control group. Statg with a regitry of the exposed individual from 1983, we updated the adesses of exposed individuals and identified a control group matched for age, sex, and neighborhood in 1979. In 1993, individual 30 years of age or older were interviewed by telephone. We obtained usable information from 795 eposed subjects and 693 control subjects.Lifetime prevalence of chloracne, abnormal nails, h s, skin allergy, goiter, headache, gum pigientation and broken teeth were observed more frquenty in the PCBIPCDF-eposed. men and women. The xpos men reported anemia 2.3 times more frequenty than contrls. The exposd men reported arthsitis and heriated intervertebral disks 4.1 and 2.9 times, respectively, more frequendy than controls. There was no dierence i reportd prevalences of other medical conditions. We condude that Taiwanese people exposed to high levels of PCBs and PCDFs reported more frequent medical problems, icuding skin diseas, goiter, anemia, and joint and spine diseases.

Med., 2007, 212 (2), [151][152][153][154][155][156][157] Polychlorinated biphenyls (PCBs) are environmental contaminants that have adverse effects on the endocrine and nervous systems. As they are still detected in breast milk and adipose tissue in humans, the accumulated PCBs may transfer from mothers to children and damage central nervous system. It is revealed from epidemiological studies that cognitive and motor functions were damaged in children born to mothers who ingested PCBs-contaminated foods. However, it remains unclear whether prenatal exposure to PCBs affects emotionality. In the present study, we therefore examined the effect of prenatal exposure to 3,3′,4, 4′,5-pentachlorobiphenyl (PCB126) on emotionality in rats by focusing on anxiogenic behavior and response of the hypothalamus-pituitary-adrenal axis to stress. Pregnant rats were treated orally with PCB126 at a dose of 30 μ g/kg or corn oil, its vehicle, on gestational day 15, and their male offspring were subjected to the following experiments at 4-5 weeks old. In an open field test, rats with prenatal exposure to PCB126 showed anxiogenic behavioral responses, including decrease in time spent in the center of an open field and the number of rearings and extension of grooming duration. Interactive behavior, which is an index of anxiety level, was shortened in the social interaction test. The increase in the serum corticosterone level induced by forced swim stress was facilitated by prenatal exposure to PCB126. This evidence suggests that PCB126 may exert anxiogenicity on the offspring of exposed dams, and dysfunction of the hypothalamus-pituitary-adrenal axis may at least in part contribute to this abnormality.3,3′,4,4′,5-pentachlorobiphenyl (PCB126); prenatal exposure; anxiogenic; corticosterone
