Polyacrylamide-Based Biocompatible Nanoplatform Enhances the Tumor Uptake, PET/fluorescence Imaging and Anticancer Activity of a Chlorophyll Analog

Abstract: In this report we demonstrate the outstanding advantages of multifunctional nanoplatforms for cancer-imaging and therapy. The non-toxic polyacrylamide (PAA) nanoparticles (size:18-25 nm) formulation drastically changed the pharmacokinetic profile of the 124I- labeled chlorophyll-a derivative (formulated in 10% ethanol in PBS) with a remarkable enhancement in tumor uptake, and significantly reduced uptake in spleen and liver. Among the various nanoformulations investigated, the 124I- labeled photosensitizer (do… Show more

“…PAAbased biodegradable and biocompatible multifunctional nanoplatform for cancerimaging and therapy containing both radioactive (I124) and nonradioactive chlorophylla based compound in desired imaging and PDT doses and pharmacodynamics profiles), can be used for dual imaging (PET/fluorescence) and PDT. Interestingly, the postloading of the iodinated analogs (I124 labeled and the corresponding nonradioactive compound) in polyacrylamidebased (PAA) nanoparticles (NPs) made a remarkable difference in in vivo biodistribution of the imaging and therapy agents with a significant increase in tumoruptake with reduced uptake in other organs, especially in spleen [100].…”

“…However, it is necessary to find a fluorescein carrier with good biocompatibility [3]. The polyacrylamide NPs are an important water-soluble polymeric material, which can form a stable colloidal dispersion system in water, showing excellent biocompatibility [4]. Furthermore, in the cross-link process of using the carrier to construct the fluorescence probe, which usually affects the intrinsic properties of fluorescent dyes and carriers.…”

“…Cancer is a leading cause of death in the US, with treatment mostly limited to non-selective methods, such as chemo, radio therapy, and surgery [1]. This has led to an expansive interest in researching selective methods of therapy to increase survival rates and general quality of life.…”
