Poly-L-arginine-Induced Internalization of Tight Junction Proteins Increases the Paracellular Permeability of the Caco-2 Cell Monolayer to Hydrophilic Macromolecules

Abstract: We investigated whether poly-l-arginine (PLA) enhances the paracellular permeability of the Caco-2 monolayer to hydrophilic macromolecules and clarified the disposition of tight junction (TJ) proteins. The transepithelial electrical resistance (TEER) and fluorescein isothiocyanate (FITC)-dextran (FD-4) permeation were determined after treatment with PLA. TJ proteins were visualized using immunofluorescence microscopy after PLA exposure and depletion, and their expression levels were determined. The barrier fun… Show more

“…Each G t (membrane conductance) was expressed in the ratio of 120 to 0 min values because the permeation-enhancing effects of HPBAAs were considered to have reached a steady state [ 7 , 11 ]. G t and the apparent permeability coefficient ( P app ) of fluorescein isothiocyanate dextran (FD-4, MW 3.4 kDa) in Caco-2 cells increased dose-dependently by application of PLO and PEG-PLO.…”

“…The effect of PEGylation on the permeation-enhancing mechanisms of HPBAAs was evaluated by immunofluorescent staining of TJ proteins. The fluorescence intensities of TJ proteins at the intercellular space decreased after exposure to PLO, suggesting that the mechanism of hydrophilic macromolecule permeation enhancement by PLO was a result of opening of the paracellular pathway due to the changing localization of TJ proteins, like PLA [ 6 , 7 ]. However, the fluorescence intensity of claudin-4 remained high even after exposure to PLO.…”
