Poly(ethylene glycol)-block-poly(l-lysine) Dendrimer:  Novel Linear Polymer/Dendrimer Block Copolymer Forming a Spherical Water-Soluble Polyionic Complex with DNA

Abstract: Methoxypoly(ethylene glycol)-block-poly(L-lysine) dendrimer was designed to form a water-soluble complex with plasmid DNA. The copolymer was synthesized by the liquid-phase peptide synthesis method. It was characterized by 1H NMR and matrix-assisted laser desorption ionization-time-of-flight (MALDI-TOF) mass spectrum. Agarose gel electrophoresis and DNase I protection assay proved that this linear polymer/dendrimer block copolymer assembled spontaneously with plasmid DNA, forming a water-soluble complex which … Show more

“…2 Another research group also reported the efficient transfer of genetic material using PAMAM dendrimers. 3,4 More recently, Choi et al 5 reported a novel linear-dendritic block copolymer that formed a water-soluble complex with plasmid DNA under physiological conditions. The complex was found to take a globular shape with a relatively narrow size distribution, and this novel copolymer may be used for gene delivery.…”

“…For example, dendritic poly(l-lysine) modified by various ligands, such as a sugar chain, endosome-disrupting agents, and an intracellular targeting signal, should be readily synthesizable by selecting several protecting groups of the lysine, such as the Boc, Z, and Fmoc groups. As candidates for the gene carrier among several types of dendritic poly(l-lysine)s, block copolymers consisting of poly(ethylene glycol) and poly(llysine) dendrimer were tested by Choi et al (1999Choi et al ( , 2000. The copolymers were 50-150 nm and formed a spherical complex with DNA; they did not show any cytotoxity toward NIH3T3 cells even at higher concentrations.…”

“…Alternatively synthetic polymers and nano particles are applied in gene delivery in recent years because of their potential advantages of low immunogenicity (49,50). Poly (L-Lysine) (PLL) has been widely used as a gene delivery carrier because its excellent nuclei acid condensation property and efficient protection of nuclei acid from the attack of nucleases (49,(51)(52)(53). We used synthesized PLL (MW 40,000) to deliver anti-mir-RNA-10b molecules on the breast cancer cell MDA-MB231.…”
