The present study was undertaken to determine a holistic picture of pollution comprising the physicochemical and microbiological properties of the Halda River water of Bangladesh. A total 45 samples were collected from five sampling sites. Different water quality parameters including temperature, pH, dissolved oxygen (DO), biological oxygen demand (BOD), chemical oxygen demand (COD), total suspended solids (TSS), total dissolved solids (TDS), total solids (TS), turbidity, concentration of heavy metals, total bacterial count (TBC), total coliform (TC), and fecal coliforms (FC) were assessed. Isolation and identification of 10 bacterial isolates was done using their cultural, morphological, physiological, biochemical characteristics, and 16S rRNA gene sequencing analysis. Antibiotic sensitivity of selected isolates was also investigated. DO (6–14.67 mgL−1), BOD (2–8 mgL−1), COD (40.43–107.63 mgL−1), TSS (818.33–3,180 mgL−1), TS (1,039.33–3,897.33 mgL−1), and turbidity (40.09–355 NTU), TBC (7.2 × 103–3.83 × 105 CFU/mL), FC (0.7 × 102–3.9 × 103/100 mL) were measured in both non breeding and breeding time that implies the pollution of the Halda river water. Heavy metal concentration was found within standard limit. Among the isolates, Bacillus wiedmannii strain HSA2 (accession no. MT102620) showed multidrug-resistant, which is a matter of great concern. According to the study findings, the water quality of the Halda river is being deteriorated, posing a threat to public health as well as the aquatic environment.

River water is very much important for domestic, agriculture and industrial use in Bangladesh which is in critical condition from long time based on research data. During last 40 years, extreme pollution events occurred in peripheral rivers surrounding Dhaka city and Karnaphuli River in Chittagong city. Present data showed that other urban rivers are also in critical condition especially Korotoa, Teesta, Rupsha, Pashur and Padma. The pollutants flowing with water made a severe pollution in downstream areas of rivers. Metals concentrations in river water was found to be higher in dry season. Dissolve oxygen (DO) was nearly zero in Buriganga River and several points in Turag, Balu, Sitalakhya and Karnaphuli River. NO 3 - , NO 2 - and PO 4 3- pollution occurred in different rivers. Zn, Cu, Fe, Pb, Cd, Ni, Mn, As and Cr concentration was above drinking water standard in most of the river and some metals was even above irrigation standard in water from several rivers. Sediment data showed very much higher metal concentrations in most of the rivers especially peripheral rivers in Dhaka and Karnaphuli, Korotoa, Teesta, Rupsha and Meghna River. Metal concentrations in sediment was above US EPA threshold value in most of the rivers. Metal concentrations in fish and agricultural crops showed that bioaccumulations of metals had occurred. The Halda River and Haor basins are considered unique breeding habitats for carps in Bangladesh. The purpose of the study was to evaluate the natural spawning environment of carp species, with an emphasis on the hydrological conditions of the Halda River, the Surma River, and Tanguar Haor. This study, which covers the years 2021 and 2022, considered two spawning seasons. The results revealed that temperature, total dissolved solids (TDS), salinity, turbidity, conductivity, and pH were significantly different in the Halda River to the Surma River and Tanguar Haor. However, sudden rainfall and upstream runoff in 2021 were deemed as major factors causing an increment in TDS and electrical conductivity. The water pollution index was excellent in the Surma River and Tanguar Haor, and good in the Halda River. The study reported moderately reduced IMC fry production in 2021 because of inconsistent turbidity, high conductivity, and increased salinity which interrupted the ideal spawning habitat. However, in 2022, the spawning environment was favorable in the Halda River, while the Surma River and Tanguar Haor were considered to be comparatively favorable in both years. Differences were also found in the climatic and meteorological conditions, which revealed uneven rainfall, higher temperatures, decreasing water discharge, and low water levels. The meteorological data also revealed that the overall rainfall showed a decreasing trend for all sites in the last 20 years causing an interrupted water discharge. It was discovered that the amount of carp spawn obtained in the Halda River was greater than that obtained in the Surma River and Tanguar Haor. In terms of hydrological conditions and spawning performance, the Surma River and Tanguar Haor were reported to have significant differences, which may be due to their geographical location.
