The biological response of periphyton chronically exposed to metals of urban origin (Cd, Ni and Zn) was investigated with a Pollution-Induced Community Tolerance (PICT) approach using a previously developed short-term toxicity test based on β-glucosidase (heterotrophic) activity. Periphyton was grown on plastic membranes immersed in indoor aquaria contaminated with metals at realistic contamination levels (0.3, 3 μg/l for Cd, 5, 50 μg/l for Ni, 20, 200 μg/l for Zn). After 3 weeks of exposure, biofilms' parameters (dry-weight, chlorophyll a concentration, heterotrophic activity) were analyzed and tolerance acquisition of the heterotrophic communities was assessed using the toxicity test. Modifications of bacterial and eukaryotic community structure were assessed with Automated Ribosomal Intergenic Spacer Analysis (ARISA). Effects of metal exposure were observed on biofilms parameters in the Cd and Zn experiments. Tolerance levels increased for both Cd-exposed biofilms, and for the high metal treatment biofilms in the Ni and Zn experiments. Analysis of the ARISA profiles showed that metal exposure affected the structure of both bacterial and eukaryotic communities. Moreover, Cd tolerance of the Zn-exposed heterotrophic communities was evaluated, which showed that the Zn-tolerant community (high metal treatment in the Zn experiment) also became tolerant to Cd (co-tolerance). The study shows that tolerance acquisition can be detected after exposure to environmental metal concentrations using β-glucosidase activity as an endpoint in short-term toxicity tests.

Floodplain lakes in the Rhine-Meuse delta of the Netherlands vary considerably in levels of sedimentbound toxicants. Microcosm experiments were done to compare the ecological impact of the fungicide triphenyltin acetate (TPT) between test systems with clean or polluted sediments (10 microcosms each). Differences in sediment quality affected the structure of the aquatic communities that developed in the microcosms. Initially, a faster growth of the macrophyte Elodea nuttallii was observed on the polluted sediments, which contained not only toxicants but also higher organic matter and nutrient levels. Dynamics of TPT concentrations in the overlying water were very similar between the two types of test system. Higher levels of TPT, however, were found in the sediment compartment of the clean sediment systems containing a smaller macrophyte biomass. TPT was very persistent in the sediments. In both test systems representatives of several taxonomic groups showed clear responses to a single application of TPT, although benthic Nematoda were not affected. Although a few differences in the intensity and/or duration of TPT-related population responses were observed between the two types of test system, the background pollutants in the polluted sediment hardly affected the overall sensitivity of the aquatic community to the additional chemical stressor TPT.

Pour caractériser l'état d'un écosystème aquatique contaminé par des toxiques il est nécessaire de développer des méthodes d'évaluation des impacts écotoxicologiques, basées à la fois sur l'étude de réponses physiologiques et sur la structure des communautés vivant dans cet écosystème. La méthode PICT (Pollution-Induced Community Tolerance) proposée par Blanck et al. (1988), répond à ces deux critères. Elle est fondée sur l'hypothèse qu'une communauté biologique naturelle est constituée de différents «composants» ayant des sensibilités variables vis-à-vis d'un toxique donné. Ainsi, suite à une exposition à un toxique, les organismes les plus sensibles ne sont plus concurrentiels et sont remplacés par des organismes plus tolérants. La communauté résultante présente alors une tolérance vis-à-vis du toxique, supérieure à celle observée pour une communauté semblable mais n'ayant pas connu de pression de sélection par ce toxique. Cette différence de réponse peut donner une indication sur l'état de pollution des milieux dans lesquels ont été échantillonnées différentes communautés. Le choix du test physiologique pour évaluer la tolérance de la communauté est fondamental car la réponse dépend d'une part, du mode d'action du toxique et d'autre part, des mécanismes à l'origine de la tolérance de la communauté. Il existe peu d'outils de bioévaluation des milieux naturels soumis à des contaminations toxiques, et cette méthode mérite donc d'être développée. Des travaux appliquant la méthode PICT à des communautés variées et dans divers milieux, permettraient de valider la méthode et de résoudre certains problèmes, quant à la détection de la tolérance. Le développement de méthodologies nouvelles pour caractériser la structure et la tolérance des communautés étudiées est fondamental pour progresser dans nos connaissances sur le risque écotoxicologique.Application of the Pollution-Induced Community Tolerance (PICT) method to algal communities: its values as a diagnostic tool for ecotoxicological risk assessment in the aquatic environment
