Poliovirus cre (2C)-Dependent Synthesis of VPgpUpU Is Required for Positive- but Not Negative-Strand RNA Synthesis

Abstract: The cre(2C) hairpin is a cis-acting replication element in poliovirus RNA and serves as a template for the synthesis of VPgpUpU. We investigated the role of the cre(2C) hairpin on VPgpUpU synthesis and viral RNA replication in preinitiation RNA replication complexes isolated from HeLa S10 translation-RNA replication reactions. cre(2C) hairpin mutations that block VPgpUpU synthesis in reconstituted assays with purified VPg and poliovirus polymerase were also found to completely inhibit VPgpUpU synthesis in prei… Show more

“…A PV 3Dpol derivative that contains a GSSG-H 6 extension at the carboxyl terminus is as active as unmodified 3Dpol in RNA-primed synthesis (51) but completely lacks any VPg uridylylation activity. 8 Only two residues of the fingers subdomain, Lys-133 and Gln-134, contribute directly to the interface.…”

“…Because both plus-and minus-strand RNAs are VPg-linked in vivo (21) it is generally accepted that VPgpUpU is the primer used by PV 3D pol for the synthesis of both RNA strands. Recent studies suggest that the VPgpUpU made on the cre(2C) is used only for plus but not for minus strand RNA synthesis (22,23). It remains to be shown how these precursors are translocated to the 3Ј end of minus strands before plus-strand RNA synthesis.…”

“…It remains to be shown how these precursors are translocated to the 3Ј end of minus strands before plus-strand RNA synthesis. Although it is also not yet known where the VPg-linked precursors are made for minus-strand synthesis, the poly(A) tail has been proposed as a possible site (9,22,23).…”
