The crosstalk between viral infections, amyloid formation and neurodegeneration has been discussed with varying intensity since the last century. Several viral proteins are known to be amyloidogenic. Post-acute sequalae (PAS) of viral infections is known for several viruses. SARS-CoV-2 and COVID-19 implicate connections between amyloid formation and severe outcomes in the acute infection, PAS and neurodegenerative diseases. Is the amyloid connection causation or just correlation? In this review we highlight several aspects where amyloids and viruses meet. The evolutionary driving forces that dictate protein amyloid formation propensity are different for viruses compared to prokaryotes and eukaryotes, while posttranslational endoproteolysis appears to be a common mechanism leading up to amyloid formation for both viral and human proteins. Not only do human and viral proteins form amyloid irrespective of each other but there are also several examples of co-operativity between amyloids, viruses and the inter-, and intra-host spread of the respective entity. Abnormal blood clotting in severe and long COVID and as a side effect in some vaccine recipients has been connected to amyloid formation of both the human fibrin and the viral Spike-protein. We conclude that there are many intersects between viruses and amyloids and, consequently, amyloid and virus research need to join forces here. We emphasize the need to accelerate development and implementation in clinical practice of antiviral drugs to preclude PAS and downstream neurological damage. There is also an ample need for retake on suitable antigen targets for the further development of next generation of vaccines against the current and coming pandemics.

La poliomielitis, causada por el poliovirus, ha sido un objetivo clave en la erradicación de enfermedades infecciosas a nivel global. Los programas de vacunación han desempeñado un rol crucial en la lucha contra esta enfermedad, y diferentes estrategias de inmunización han sido desarrolladas a lo largo del tiempo. Una de estas estrategias es la combinación de vacunas secuenciales inactivadas (IPV) y vivas orales (OPV) contra el poliovirus. Se llevó a cabo una revisión bibliográfica con el propósito de analizar la literatura existente sobre efectividad y seguridad de la combinación de IPV y OPV en esquemas de vacunación contra el poliovirus. La metodología empleada se sustenta en la recopilación y evaluación de estudios previos, artículos científicos y datos de ensayos clínicos relacionados con esta estrategia. Los hallazgos de esta revisión indicaron que la combinación de IPV y OPV puede ser efectiva en la protección contra el poliovirus. Esta estrategia ha demostrado aumentar la inmunogenicidad, particularmente en lo que respecta al serotipo 2 del poliovirus, y reducir los riesgos asociados con la OPV, como la polio paralítica vinculada a la vacuna (VAPP). En conclusión, la combinación de IPV y OPV en esquemas de vacunación contra el poliovirus es una estrategia prometedora que puede contribuir significativamente a la erradicación de la enfermedad. Sin embargo, es crucial mantener una supervisión constante y una evaluación rigurosa para garantizar su eficacia y seguridad a largo plazo. Esta revisión resalta la importancia de considerar esta estrategia en los programas de vacunación para alcanzar el objetivo de un mundo libre de polio de manera más efectiva.

Poliovirus (PV), the virus that causes both acute poliomyelitis and post-polio syndrome, is classified within the Enterovirus C species, and there are three wild PV serotypes: WPV1, WPV2 and WPV3. The launch of the Global Polio Eradication Initiative (GPEI) in 1988 eradicated two of the three serotypes of WPV (WPV2 and WPV3). However, the endemic transmission of WPV1 persists in Afghanistan and Pakistan in 2022. There are cases of paralytic polio due to the loss of viral attenuation in the oral poliovirus vaccine (OPV), known as vaccine-derived poliovirus (VDPV). Between January 2021 and May 2023, a total of 2141 circulating VDPV (cVDPV) cases were reported in 36 countries worldwide. Because of this risk, inactivated poliovirus (IPV) is being used more widely, and attenuated PV2 has been removed from OPV formulations to obtain bivalent OPV (containing only types 1 and 3). In order to avoid the reversion of attenuated OPV strains, the new OPV, which is more stable due to genome-wide modifications, as well as sabin IPV and virus-like particle (VLP) vaccines, is being developed and offers promising solutions for eradicating WP1 and VDPV.
