Polarization of PI3K Activity Initiated by Ooplasmic Segregation Guides Nuclear Migration in the Mesendoderm

Abstract: Asymmetric localization of RNA is a widely observed mechanism of cell polarization. Using embryos of the ascidian, Halocynthia roretzi, we previously showed that mesoderm and endoderm fates are separated by localization of mRNA encoding a transcription factor, Not, to the future mesoderm-side cytoplasm of the mesendoderm cell through asymmetric positioning of the nucleus. Here, we investigated the mechanism that defines the direction of the nuclear migration. We show that localization of PtdIns(3,4,5)P3 to the… Show more

“…Detection of nodal mRNA and the active form of Arp2/3 complex, as well as of bmp2/4 mRNA and phosphorylated Smad1 (pSmad1) was performed. Riboprobes were first detected by TSA Plus cyanine 3/fluorescein system (PerkinElmer, MA) as described previously [ 11 ]. After the completion of WISH, the specimens were subjected to immunostaining.…”

“…Ascidians also display ooplasmic dynamics more remarkably than anamniote vertebrates do, however they do not specify the dorsal fate but the posterior fate by this process [ 8 , 9 ]. Dorsal structures are induced by asymmetrical segregation of mRNAs during cleavage and Fgf signalling emanating from endodermal precursor cells [ 10 , 11 ].…”

“…Their many advantages as laboratory organisms have carried the tunicates through the centuries of experimental biology, and their status as the sister group to the vertebrates has helped secure a spot for them at the biomedical research table. Studies in tunicates have helped establish basic concepts in developmental biology such as invariant lineages and mosaic development [64, 66, 191, 352], and have shed light on transcriptional and cellular mechanisms of development [26, 59, 68, 69, 84, 101, 136, 167, 226, 234, 262, 265, 323]. Furthermore, comparative studies using tunicates have refined models of chordate and vertebrate evolution [1, 2, 9, 49, 92, 112, 156, 192, 205, 275, 311, 345].…”

“…In turn, tumor cell-produced ligands such as colony-stimulating factor 1 (CSF1) signal to CSF1R on TAMs to promote recruitment and M2-like polarization 41 , 43 . Downstream PI3K signaling is implicated in such signaling and MT dynamics 44 , 45 , and in TAMs, reports show PI3K inhibition (particularly of isoform p110γ) reprograms cells toward M1-like polarization 46 .…”
