Pneumocandins from Zalerion arboricola. I. Discovery and isolation.

Abstract: HPLCbioautography of the directed biosynthesis of Zalerion arboricola led to the discovery of pneumocandin Bo (L-688,786), a new antifungal and anti-Pneumocystis carinii lipopeptide. Isolation techniques were developed to separate this componentfrom pneumocandinAo(L-67 1 ,329) in fermentations of a mutant of Zalerion arboricola. Anumberof related compounds were also isolated, which differ from pneumocandinsAoand Bo in the hydroxylation patterns on the ornithine, homotyrosine, and proline. Aculeacin1 '2) and ec… Show more

“…These compounds belong to the echinocandin family, a class of antifungal agents that act on the fungal cell wall by inhibiting glucan synthesis. The members of the echinocandin family of fungal origin are FR901379 [14,15], echinocandin B [16], aclueacins [17], mulundocandin [18] and pneumocandins [19], which are produced by C. empetri No. 11899, Aspergillus nidulans, A. aculeatus, A. sydowi and Glarea lozoyensis, respectively.…”

“…91-75 [35], and the helioferins, protonophoric antibiotics isolated from Mycogone rosea that act by disturbing cellular ion flux and energy metabolism [36]. Although these lipopeptides have a shorter fatty acyl component (C8-C,,), they do contain alanine and isoleucine [35] and phenylalanine [36], and the fatty acyl unit is connected via an amide linkage [35, 361. Other examples include the pneumocandins [37], lipopeptides that contain a cyclic peptide made up of six amino acids (including threonine and various unusual amino acids) that are in an amide linkage to a 10,12-dimethylmyristic acid residue [38]. Methylated amino acids have also been observed in other lipopeptides, including some of the pneumocandins, [38] and enopeptin A, another lipopeptide antibiotic isolated from Streptomyces griseus [39].…”

“…arboricola (ATCC20957) was carried out as described by Schwartz et al (1992). 1 0) Isolation and Purification of Pneumocandin Do (9) The MeOHextract of the whole broth from a large scale fermentation of Z. arboricola (ATCC20957) (1.5 : 1 whole broth-MeOHratio; 10,750 liters whole broth in total) was centrifuged, adjusted with water to less than 50% MeOH(Karl Fischer analysis) and adsorbed onto a fluidized bed of SP-207 (600 liters) in an up flow direction.…”
