Allergenic pollen is produced by the flowers of a number of trees, grasses and weeds found throughout the UK. Exposure to such pollen grains can exacerbate pollen-related asthma and allergenic conditions such as allergic rhinitis (hay fever). Maps showing the location of these allergenic taxa have many applications: they can be used to provide advice on risk assessments; combined with health data to inform research on health impacts such as respiratory hospital admissions; combined with weather data to improve pollen forecasting systems; or as inputs to pollen emission models. In this study we present 1 km resolution maps of 12 taxa of trees, grass and weeds found in the UK. We have selected the main species recorded by the UK pollen network. The taxa mapped in this study were: Alnus (alder), Fraxinus (ash), Betula (birch), Corylus (hazel), Quercus (oak), Pinus (pine) and Salix (willow), Poaceae (grass), Artemisia (mugwort), Plantago (plantain), Rumex (dock, sorrels) and Urtica (nettle). We also focus on one high population centre and present maps showing local level detail around the city of London. Our results show the different geographical distributions of the 12 taxa of trees, weeds and grass, which can be used to study plants in the UK associated with allergy and allergic asthma. These maps have been produced in order to study environmental exposure and human health, although there are many possible applications. This novel method not only provides maps of many different plant types, but also at high resolution across regions of the UK, and we uniquely present 12 key plant taxa using a consistent methodology. To consider the impact on human health due to exposure of the pollen grains, it is important to consider the timing of pollen release, and its dispersal, as well as the effect on air quality, which is also discussed here.

Flow cytometry is a method widely used to quantify suspended solids such as cells or bacteria in a size range from 0.5 to several tens of micrometers in diameter. In addition to a characterization of forward and sideward scatter properties, it enables the use of fluorescent labeled markers like antibodies to detect respective structures. Using indirect antibody staining, flow cytometry is employed here to quantify birch pollen allergen (precisely Bet v 1)-loaded particles of 0.5 to 10 µm in diameter in inhalable particulate matter (PM10, particle size ≤10 µm in diameter). PM10 particles may act as carriers of adsorbed allergens possibly transporting them to the lower respiratory tract, where they could trigger allergic reactions.So far the allergen content of PM10 has been studied by means of enzyme linked immunosorbent assays (ELISAs) and scanning electron microscopy. ELISA measures the dissolved and not the particle-bound allergen. Compared to scanning electron microscopy, which can visualize allergen-loaded particles, flow cytometry may additionally quantify them. As allergen content of ambient air can deviate from birch pollen count, allergic symptoms might perhaps correlate better with allergen exposure than with pollen count. In conjunction with clinical data, the presented method offers the opportunity to test in future experiments whether allergic reactions to birch pollen antigens are associated with the Bet v 1 allergen content of PM10 particles >0.5 µm.

BACKGROUND Pollen allergies concern a significant proportion of the population globally. Today, online tools such as pollen diaries and mobile applications allow an easy and fast documentation of allergic symptoms via the internet. OBJECTIVE This study aimed to characterize the users of the Patient´s Hayfever Diary, to apply different symptom score calculations for comparison and to evaluate the contribution of organs and medication to the total score for the first time. METHODS Users of the Patient´s Hayfever Diary were filtered concerning their location (Austria/Germany), significant positive correlation to the respective pollen type (birch/grass) and at least 15 entries in the respective season. Four different symptom score calculation methods were applied to the datasets from 2009 until 2018, among them two raw symptom scores and two symptom load indices (normalized) calculations. Pearson correlation coefficients were calculated pairwise for four symptom score calculations. RESULTS Users are mostly male and in the age group of 21 to 40 years or in the group above 40 years. User numbers increased in the last years, especially when mobile applications were made available. The Pearson correlation coefficients show a significant linear relationship above 0.9 between the four symptom score datasets. The nose contributes the most to the symptom score and determines about 40% of the score. CONCLUSIONS The exact method of calculation of the symptom score is not critical. All computation methods show the same behavior (increase/decrease). The symptom load index is therefore a useful computation method in all fields exploring pollen allergy and online diaries are a globally applicable tool to monitor the effect of pollen on human health via electronic generated symptom data. However, studies should always consider the variation between different pollen seasons, years and biogeographical regions.
