Pluronic F‐127: An Efficient Delivery Vehicle for 3‐(1'‐hexyloxy)ethyl‐3‐devinylpyropheophorbide‐a (HPPH or Photochlor)

Abstract: To determine the impact of delivery vehicles in photosensitizing efficacy of HPPH, a hydrophobic photosensitizer was dissolved in various formulations: 1% Tween 80/5% dextrose, Pluronic P‐123 and Pluronic F‐127 in 0.5%, 1% and 2% phosphate buffer solutions (PBS). HPPH was also conjugated to Pluronic F‐127, and the resulting conjugate (PL‐20) was formulated in PBS. Among the different delivery vehicles, only Pluronic P‐123 displayed significant vehicle cytotoxicity, whereas Pluronic F127 was nontoxic. Compared … Show more

“…S1C) determined using dynamic light scattering was 30.4 nm (±2.7 nm), and the zeta potential was +5.82 mV (±0.89 mV) with a critical micellar concentration (CMC) of about 0.3 μg/ml. In contrast, the mean diameter of Pluronic F127 micelles was 27.1 (±3.2 nm) and the zeta potential was −0.30 mV (±0.17 mV), consistent with literature values ( 24 , 25 ).…”

“…(h) Impact of Pluronic F-127 formulation in photophysical properties of PS 1: Similar to most of the tetrapyrrole-based PS, e.g., HPPH 30 , the PS 1 derived from chlorophyll-a, on formulating in Pluronic F-127/PBS solution forms aggregation, which significantly reduces its absorption and fluorescence intensities. However, in the presence of HSA (human serum albumin) or BSA (bovine serum albumin), the PS self-aggregation disaggregates, and exhibit photophysical properties similar to the respective monomers observed in organic solvents (e.g., methanol or tetrahydrofuran).…”

“…This report presents (a) the utility of a corresponding non-labeled iodinated PS 1 for treating lung cancer with and without chemotherapy (doxorubicin) in a variety of lung tumors xenografts derived from lung cancer patients and (b) the toxicity and toxicokinetic profiles of the PS formulated in Pluronic F-127 at variable doses in male and female rats and dogs. We and others have previously shown the improved PDT efficacy of certain tetrapyrrolic photosensitizers in a Pluronic-based formulation either by encapsulation or by conjugating the PS with Pluronic F-127 with and without the combination of co-delivery of doxorubicin for overcoming drug resistance in cancer [29][30][31].…”

“…In addition, Pluronics â are characterized by their amphiphilic structure (molecules that have both hydrophilic and hydrophobic regions) and they are able to selfaggregate in aqueous solution (54,55). Several studies have reported Pluronics â as an efficient drug delivery system for cancer treatment and other applications (56)(57)(58)(59)(60).…”
