PLETHORA transcription factors orchestrate de novo organ patterning during Arabidopsis lateral root outgrowth

Abstract: SignificanceRoot architecture is an important trait that is shaped by the formation of primary roots, lateral roots, and adventitious roots. Here, we show that three PLETHORA (PLT) transcription factors are the key molecular triggers for the de novo organ patterning during Arabidopsis lateral root formation. PLT3, PLT5, and PLT7 redundantly regulate the correct initiation of formative cell divisions in incipient lateral root primordia and the proper establishment of gene expression programs that lead to the fo… Show more

“…4B). PLT3, TCP20, and TCP21 fusion proteins were detected at stage I, whereas SCR expression was consistently observed in the stage II outer cell layer (Goh et al 2016;Du and Scheres 2017). Notably, WOX5 promoter activity was detected from stage II onward in cells where PLT, SCR, and TCP proteins are all expressed.…”

During lateral root initiation, two to three adjacent pericycle cell files undergo several rounds of anticlinal divisions to create a single layered stage I lateral root primordium (LRP) composed of up to eight to 10 cells of equal size.…”

“…We, therefore, next, investigated the genes regulated by OsWOX10 during CRP initiation and outgrowth. In rice, OsERF3 and OsCRL1 regulate CRP initiation, whereas Arabidopsis PLT genes functions during LR primordia outgrowth Zhao et al, 2015;Du and Scheres, 2017). Consistently, the expression of OsERF3 and OsCRL1 and their related genes (i.e.…”

“…During the course of LR development, specific modules of AUXIN RESPONSE FACTOR (ARF) transcription factors and AUXIN/INDOLE ACETIC ACID (AUX/IAA) repressor proteins, as well as other downstream transcription factors, specify LR primordium (LRP), founder cells and promote LR formation [37][38][39]. In parallel, three PLETHORA transcription family members-PLT3, PLT5, and PLT7-contribute to the establishment of LRPs [40]. During LR development, quiescent center (QC) is established by the action of a transcription factor complex comprising PLT, SCARECROW (SCR), and several members of TEOSINTE BRANCHED1/CYCLOIDEA/PCF (TCP) through activation of WUSCHEL-RELATED HOMEOBOX5 (WOX5) expression [41,42].…”
